TNRC18在结直肠癌组织中的表达及临床病理意义

Expression of TNRC18 in colorectal cancer and its clinicopathological significance

目的:探讨TNRC18在结直肠癌组织中的表达与临床病理参数的相关性及意义。方法:应用组织芯片技术将搜集的蜡块组织进行整理做成组织芯片,从UCSC xena网站(http://xena.ucsc.edu./)下载结直肠癌(TCGA-COAD)数据库,用统计学方法分析结直肠癌中TNRC18 mRNA表达与临床病理参数的相关性。采用免疫组化EnVision两步法分别检测结直肠癌和癌旁正常组织中TNRC18的表达,并分析其与结直肠癌临床病理参数的相关性。结果:TNRC18蛋白在结直肠癌和癌旁正常组织细胞核中的阳性率为90.75%、75.72%,差异具有统计学意义(P<0.05)。TNRC18蛋白在结直肠癌和癌旁正常组织细胞浆中的阳性率为90.17%、0.58%,差异具有统计学意义(P<0.05)。TNRC18在mRNA水平和结直肠癌组织细胞浆中的表达与淋巴结转移(P<0.05)和TNM分期有关(P<0.05),而与其他临床病理参数尚无相关性(P>0.05)。结论:TNRC18在结直肠癌中的表达升高,可能作为检测结直肠癌进展和转移的潜在分子标志物。

Objective:To explore the relationship and significance of trinucleotide repeat containing 18 protein(TNRC18)expression in colorectal cancer(CRC)with clinicopathological parameters.Methods:Tissue chip technology was used to organize the collected wax blocks into tissue microarrays.The colorectal cancer database(TCGA-COAD)was downloaded from the UCSC xena website(http://xena.ucsc.edu./),and the relationship between TNRC18 mRNA expression and the clinicopathological parameters was analyzed statistically.The TNRC18 expression in CRC and adjacent tissues was examined by immunohistochemistry,and its correlation with clinical pathological parameters of CRC was analyzed.Results:The positive rate of TNRC18 protein in the nucleus of CRC and adjacent tissues was 90.75%and 75.72%,and the difference was statistically significant(P<0.05).The positive rate of TNRC18 protein in the cytoplasm of CRC and adjacent tissues was 90.17%and 0.58%,and the difference was statistically significant(P<0.05).The expression of TNRC18 in mRNA and colorectal cancer cytoplasm was related to lymph nodes metastasis(P<0.05)and TNM stages(P<0.05),but not related to other clinicopathological features(P>0.05).Conclusion:TNRC18 was high expression in CRC,which may be a potential molecular marker for detecting the progression and metastasis of CRC.
