广藿香酮传递体包封率测定方法的筛选研究 被引量:8

摘要 目的考察超速离心法、超滤离心法、鱼精蛋白沉淀法和微柱离心法用于测定传递体包封率的可行性,并从中优选出适用于测定广藿香酮传递体包封率的最佳方法。方法采用薄膜分散法制备空白传递体及广藿香酮传递体;建立广藿香酮HPLC含量测定方法;采用4种方法分离游离广藿香酮和传递体,并进行样品包封率测定,以回收率和分离度为指标评价方法的适用性。结果在广藿香酮和传递体混合物分离试验中,除超速离心法外,其余3种方法均能较好地分离游离广藿香酮和传递体。间接测定包封率的超滤离心法和鱼精蛋白沉淀法广藿香酮回收率分别为93.8%、103.0%,RSD分别为1.0%、0.80%。在广藿香酮传递体样品包封率测定试验中,4种方法测得的包封率分别为41.83%、46.79%、44.40%、4.24%,其RSD分别为5.4%、4.0%、0.14%、5.4%。结论鱼精蛋白沉淀法操作简单,分离度、重复性较好,适用于广藿香酮传递体包封率的测定。 Objective To determine the feasibility of ultracentrifugation method,ultrafiltration centrifugation method,protamine precipitation method,and microcolumn centrifugation method for the determination of encapsulation efficiency of transfersomes,and to select the best method to determine the encapsulation efficiency of pogostone transfersomes.Methods Pogostone and blank transfersomes samples were prepared by film dispersion method.A HPLC method for the determination of pogostone was established.The mixture of pogostone and transfersomes was separated by ultracentrifugation method,ultrafiltration centrifugation method,protamine precipitation method,and microcolumn centrifugation method,which were used to determine the encapsulation efficiency of pogostone transfersomes samples.The adaptability of the method was evaluated by the recovery and separation.Results In the separation of pogostone and transfersomes mixtures,except for the ultracentrifugation method,the remaining three methods all well separated the physical mixture of pogostone and transfersomes.The recoveries of free pogostone were 93.8%and 103.0%,and the RSDs were 1.0%and 0.80%respectively,with ultrafiltration centrifugation method and protamine precipitation methodthe indirect measure of encapsulation efficiency.In the determination of encapsulation efficiency of pogostone transfersome samples,the encapsulation efficiencies by the four methods were 41.83%,46.79%,44.40%,and 4.24%respectively,and the RSDs were 5.4%,4.0%,0.14%,and 5.4%respectively.Conclusion The protamine precipitation method is simple,stable and repeatable,and is the best choice in the determination of encapsulation efficiency of pogostone transfersomes.
