益脉颗粒治疗动脉粥样硬化的实验研究

摘要 目的探讨益脉颗粒对动脉粥样硬化(AS)兔血脂、动脉硬化程度、植物血凝素样氧化低密度脂蛋白受体1(LOX-1)表达的影响。方法按照既定实验方法测定各组兔血脂(总胆固醇、低密度脂蛋白胆固醇),HE染色做病理学观察,免疫组化测定LOX-1表达水平,实时定量PCR反应检测基因表达。结果与正常对照组相比,高脂组TC(总胆固醇)、LDL-c(低密度脂蛋白胆固醇)水平均明显高于中药组;血管病理学观察,正常对照组在肉眼观察下未发现AS斑块,在镜下可以见到完整的内膜;中药组与高脂组相比,斑块的面积减少,内膜增厚。中药组LOX-1的蛋白分布较稀疏,高脂组LOX-1的蛋白分布较密集;中药组LOX-1基因表达低于高脂组。结论益脉颗粒可以明显地抑制LOX-1蛋白表达和基因表达,改善血管内皮功能,延缓动脉粥样硬化进程。 Objective To study the mechanism of action of Yimai granule in resistance to atherosclerosis by establishing rabbits models to perform experiments.Methods We took chest aortas after executing rabbits and used HE staining to observe atherosclerosis.At the same time,we used immunohistochemical staining to observe LOX-1 protein expressions and used RT-PCR to investigate gene expressions.Results Compared with the normal control group,the levels of TC(total cholesterol)and LDL-c(low density lipoprotein cholesterol)in the hyperlipidemia group were significantly higher than those in the traditional Chinese medicine group;vascular pathology observation showed that no AS plaques were found in the normal control group,and complete intima could be seen under microscope.Compared with the hyperlipidemia group,the area of plaques in the traditional Chinese medicine group decreased and intima thickened.The protein distribution of LOX-1 in the traditional Chinese medicine group was sparse,and the protein distribution of LOX-1 in the traditional Chinese medicine group was relatively dense,and the expression of LOX-1 gene in the traditional Chinese medicine group was lower than that of the control group.Conclusion Yimai granule can significantly inhibite the protein expression and gene expression of lox-1 in AS,and protect the function of endothelial cells.
