复方雷公藤汤最佳剂量配比的正交设计对肾病大鼠心脏减毒增效作用的实验研究

Experimental Study on the Decreasing Toxicity and Increasing Synergistic Effect of Compound Tripterygium Wilfordii Decoction on Heart of Nephrotic Rats by Orthogonal Design

目的研究复方雷公藤汤正交设计对肾病大鼠心脏的减毒增效作用的最佳剂量配比。方法大鼠尾静脉注射阿霉素,复制微小病变肾病综合征模型。采用正交实验法进行设计,将肾病模型大鼠随机分为9组,每组10只,连续灌服不同剂量配比复方雷公藤汤14 d。增效测量24 h尿蛋白,对心脏减毒测量肌酸激酶、肌酸激酶同工酶。筛选出复方雷公藤汤最佳剂量配比。结果经正交设计的9个复方雷公藤汤不同剂量配比对肾病大鼠心脏的减毒实验结果表明:从直观分析可见,影响因素大小的排序雷公藤>甘草>黄芪>当归,其中8号复方效果最好,最优为A3B2C1D3,即雷公藤60 g,黄芪120 g,当归72 g,甘草12 g。经正交设计的9个组成的复方雷公藤汤不同剂量配比对降低肾病大鼠24 h尿蛋白实验结果表明:从直观分析可见,影响因素大小的的排序雷公藤>甘草>黄芪,当归无影响,其中3号复方效果最好,最优为A1B3C3D3,即雷公藤120 g,黄芪60 g,当归36 g,甘草12 g。结论复方雷公藤汤治疗肾病时减低心脏毒性为主,采用A3B2C1D3配比组成复方,治疗肾病时降低24 h尿蛋白为主,采用A1B3C3D3配比组成复方,二种配比复方均起到减毒增效作用。

Objective:To study the optimum dosage ratio of Compound Tripterygium Wilfordii Decoction in reducing toxicity and synergistic effect on the heart of nephrotic rats by orthogonal design.Methods:Rats were injected with doxorubicin in the tail vein to replicate a model of minimal lesion nephrotic syndrome.Rats with nephropathy were randomly divided into 9 groups,10 rats in each group,and the Compound Tripterygium Wilfordii Decoctionwas administered at different doses for 14 days.The effect was measured for 24 hour urine protein,and the creatine kinase and creatine kinase isoenzyme were measured for cardiac attenuation.The optimal dosage ratio of Compound Tripterygium Wilfordii Decoction was screened out.Results:The Results of attenuation experiments with different dosage ratios showed that the order of influencing factors was Tripterygium wilfordii>Licorice>Astragalus membranaceus>Angelica sinensis,of which compound No.8 had the best effect and the best was A3 B2 C1 D3(Tripterygium wilfordii 60 g,Astragalus membranaceus 120 g,Angelica sinensis 72 g,Licorice 12 g).The Results of 24-hour urinary protein reduction with different dosage ratios showed that the order of influencing factors was Tripterygium wilfordii>Licorice>Astragalus membranaceus,and Angelica sinensis had no effect.Compound No.3 had the best effect,and A1 B3 C3 D3(Tripterygium wilfordii 120 g,Astragalus membranaceus 60 g,Angelica sinensis 36 g,Licorice 12 g)had the best effect.Conclusion:When using Compound Tripterygium Wilfordii Decoction on nephropathy,A3 B2 C1 D3 ratio is to reduce cardiac toxicity,A1 B3 C3 D3 ratio to reduce 24-hour urinary protein mainly.Both of the two mixtures play a role in reducing toxicity and increasing synergy.
