白芍总苷对局灶性脑缺血大鼠海马CA1区病理改变的影响 被引量:2

Effect of Total Paeony Glucoside on Pathological Changes of Hippocampal CA1 Area in Rats with Focal Cerebral Ischemia

摘要 目的探讨白芍总苷对局灶性脑缺血中枢海马CA1区病理改变的影响。方法通过阻断大脑中动脉制备局灶性脑缺血大鼠模型。白芍总苷低、中、高剂量组分别每日1次灌胃给予白芍总苷50、100、200 mg/kg,疗程28 d。红四氮唑(TTC)染色法计算脑梗死体积,干湿比重法计算脑组织含水量,苏木精-伊红(HE)法进行中枢海马CA1区病理学检查,原位末端转移酶标记染色法(TUNEL)观察CA1区神经元凋亡状况并计算凋亡指数(AI),透射电子显微镜(TEM)观察CA1区神经元超微结构变化。结果白芍总苷中、高剂量组能够明显降低局灶性脑缺血大鼠脑梗死体积(P<0.05或P<0.01),降低脑组织含水量(P<0.05),明显改善中枢海马CA1区组织细胞病理性改变,抑制CA1区神经元凋亡、AI显著降低(P<0.01),明显改善CA1区神经元细胞器等超微结构病理性改变。结论白芍总苷对局灶性脑缺血中枢海马CA1区病理改变及CA1区神经元超微结构病理改变具有保护作用。 Objective:To study the effect of Total Paeony Glucoside(TPG)on pathological changes of hippocam pal CA1 area in rats with focal cerebral ischemia.Methods:The model of focal cerebral ischemic was established by blocking the middle cerebral artery.TPG low,middle and high dose groups were given intraperitoneal adminis tration 50,100,200 mg/kg,once a day for 28 days.The water content of brain tissue was calculated with dry-wet specific gravity method;the volume of cerebral infarction was calculated with red tetrazolium(TTC)staining meth od,the pathological examination of central hippocampal CA1 area by HE staining;the neuronal apoptosis in CA1 area were observed by TUNEL staining and AI were calculated;the ultrastructural changes in neurons in CA1 re gion were observed by TEM.Results:TPG in middle and high dose groups could significantly reduce cerebral in farction volume(P<0.05 or P<0.01)and brain water content(P<0.05),improve the pathological changes of hip pocampal CA1 area,inhibit neuronal apoptosis and AI(P<0.01),and improve the ultrastructural pathological changes such as neuronal organelles.Conclusion:TPG has protective effects on the pathological changes of CA1 in hippocampus of central cerebral ischemia and the ultrastructural pathological changes of neurons in CA1 area.
