穴位贴敷疗法对高尿酸血症大鼠模型血尿酸及相关酶活性的影响

Effect of Acupoint Application Therapy on SUA and the Activity of Related Enzyme in Hyperuricemia Rats

目的观察穴位贴敷疗法对高尿酸血症大鼠模型血尿酸及相关酶活性的影响并探讨其作用机制。方法将32只清洁级雄性SD大鼠随机分为空白组、模型组、凡士林贴敷组、穴位贴敷组,每组8只。采用氧嗪酸钾灌胃构建高尿酸血症大鼠模型。凡士林贴敷组运用凡士林贴敷双侧肝俞和期门、脾俞和章门、肾俞和京门,穴位贴敷组运用由柴胡、芍药、熟地黄、泽泻、茯苓、白术、白芥子等方药组成的穴位药贴贴敷相同的穴位。采用全自动生化分析仪测定血清中SUA、ALT、AST水平;酶比色法检测肝组织XOD和ADA的活性;酶联免疫法(ELISA)检测肝组织GDA和HGPRT含量。结果与空白组比较,模型组SUA、XOD和GDA含量均增高(P<0.05或P<0.01),差异有统计学意义;与模型组和凡士林贴敷组比较,穴位贴敷组SUA、XOD和GDA含量均降低(P<0.05或P<0.01),ALT和ADA含量升高(P<0.01),差异有统计学意义。结论穴位贴敷疗法可以有效降低高尿酸血症大鼠模型SUA水平,其机制可能是通过抑制肝组织中XOD和GDA活性。

Objective:To observe the effect of acupoint application on serum uric acid and related enzyme activity in hyperuricemia rats and explore its mechanism.Methods:32 clean male SD rats were randomly divided into the blank group,the model group,vaseline group and acupoint application group,8 rats in each.The hyperuricemia rat model was established by intragastric administration of potassium oxyzinate.In vaseline group,Vaseline was applied to the two sides of"Ganshu and qimen,Pishu and zhangmen,Shenshu and Jingmen".Acupoint application group applied the same acupoints with traditional Chinese medicine.The serum levels of SUA,ALT and AST were measured by automatic biochemical analyzer.The activity of XOD and ADA in liver tissue was detected by enzyme colorimetry.The contents of GDA and HGPRT were detected by ELISA.Results:Compared with the blank group,the content of SUA,XOD and GDA in the model group were all increased,with statistical difference(P<0.05 or P<0.01).Compared with the model group and vaseline group,those in acupoint application group decreased(P<0.05 or P<0.01),and the content of ALT and ADA increased,with statistical difference(P<0.01).Conclusion:Acupoint application therapy can reduce the level of SUA in rats with hyperuricemia,whose mechanism may inhibit XOD and GDA activity of liver.
