摘要 白芨(Bletilla striata)是我国重要的药用植物,组织培养技术繁育组培苗已成为当前白芨人工栽培的主要种苗来源,因此对白芨组培苗的真伪鉴定成为市场需求的重要技术。本研究为了探索鉴定白芨真伪的新方法,设计了20对SSR引物和22对ISSR引物,对1份正品白芨标准品和7份外观较相似的白芨组培苗进行分子鉴定。结果表明,多对引物检测出4~8号样品与1号标准品带型存在差异,表明此5份组培苗为假的白芨;35对引物均未检测出2号和3号样品与1号标准品带型存在差异,表明2号和3号组培苗为真的紫花白芨。本文建立的方法可作为白芨真伪鉴别的分子依据,为市场上白芨组培苗的快速鉴定提供了技术支持。 Bletilla striata is an important medicinal plant in China. The authenticity identification of tissuecultured seedlings was becoming the important technology demanded in the market since the plantlets had become the main source of artificial cultivation. In order to explore a new method for the identification of B.striata, 20 pairs of SSR primers and 22 pairs of ISSR primers were designed to amplify 1 certified B. striata plantlet sample cultured in the lab and 7 plantlets samples bought from the market. The results showed that the No.4~ No.8 samples performed differently with the No.1 standard sample detected by multiple pairs of primers, which indicated that they were false B. striata seedlings. There were no difference for the banding patterns between No.2 and No.3 samples and No. 1 standard sample, indicating that No.2 and No.3 samples were more likely to be true purple flower B. striata seedlings. The method established in this paper could provide the molecular basis and technical support for the authenticity identification of Bletilla striata.
