犬弓首蛔虫衡阳分离株的核糖体DNA ITS序列分析 被引量:6

摘要 为在初步了解衡阳市犬弓首蛔虫病的流行,并分析其遗传变异情况。应用饱和盐水漂浮法对衡阳市453份犬粪便进行犬弓首蛔虫分离,同时应用PCR技术对部分犬弓首蛔虫衡阳株核糖体ITS序列进行扩增、测序与分析。结果显示,衡阳市犬弓首蛔虫感染率为17.07%,5株犬弓首蛔虫分离株ITS序列基本一致,种内遗传差异为98.3%~99.0%,与基因库中其他蛔虫的同源性均低于41.0%。本研究通过建立NJ系统发育树,发现所获得的分离株与已知犬弓首蛔虫分离株聚为同一大分支,与其他蛔虫所属分支距离较远。研究结果表明,衡阳市犬弓首蛔虫感染情况较为严重,对人类健康造成一定的潜在威胁,需要引起相关部门的重视。本研究可为该地区犬弓首蛔虫病的防控提供依据,并为犬弓首蛔虫进一步的分类、鉴定和遗传变异研究提供参考。 The objective of the present study was to investigate the epidemiology of Toxocara canis in Hengyang area,Hunan province and phylogenic relationship of the isolated parasites.Total 453 fecal samples were collected from dogs and T.canis was isolated by saturated saline floating method.The T.canis positive rate was 17.07%among the tested dogs.The ribosome DNA internal transcribed spacer(ITS)of some isolates was amplified by PCR and sequenced for phylogenic tree analysis.The results showed the nucleotide sequence identity at 98.3–99%among the obtained isolates and belonged to the same branch in phylogenic analysis.Among the isolated parasites,5 isolates shared the same nucleotide sequences.These isolates from this study were genetically far distance from other ascaris(below 41%)available in GenBank.In conclusion,T.canis infection was prevailing in Hengyang area and the present study provided useful information on further research for the prevention and treatment of the disease.
