长链非编码RNA SOX2OT对内皮细胞生物学功能的影响

摘要 目的 探索长链非编码RNA SOX2OT(LncRNA SOX2OT)对内皮细胞(EC)增殖、细胞周期、迁移和侵袭能力的影响.方法 首先体外分离培养EC,分别采用实时荧光定量PCR(RT-PCR)检测SOX2OT在EC之间的表达差异;CCK8和集落形成实验观察EC的增殖情况;流式细胞术观察EC的细胞周期和凋亡变化;划痕愈合实验和Transwell小室实验检测EC迁移和侵袭能力.结果 SOX2OT在EC中相对表达量与对照组比较,差异有统计学意义(P<0.05).转染si-SOX2OT和si-NC到EC中,发现转染si-SOX2OT组的细胞增殖能力明显下降,差异有统计学意义(P<0.05).转染si-SOX2OT的细胞凋亡率明显增加,且细胞周期被阻滞在G0/G1期;si-SOX2OT可抑制细胞的迁移和侵袭能力.结论 LncRNA SOX2OT在EC中表达上调,抑制其表达可以抑制内皮细胞增殖、阻滞细胞周期、促进凋亡及抑制肿瘤细胞侵袭和迁移.研究SOX2OT为内皮细胞的诊治及预防转移复发等提供了重要的理论基础. Objective To explore the effects of long non-coding RNA SOX2OT(LneRNA SOX2OT)on the proliferation,cell cycle,migration and invasion of endothelial cells(EC).Methods Firstly,EC was isolated and cultured in vitro,a nd then real-time fluorescent quantitative PCR(RT-PCR)was used to detect the difference of SOX2OT expression between ECs;CCK8 and colony formation experiments were used to observe the proliferation of EC;flow cytometry was used to observe the cell cycle and apoptotic changes of EC;scratch healing experiment and Transwell chamber experiment were used to detect the migration and invasion ability of EC.Results There was significant difference on the relative expression of SOX2OT in EC between EC group(1.76±0.19)and control group(1.07±0.05)(P<0.05).When transfected si-SOX2OT and si-NC into EC,the proliferation of cells in the transfected group decreased significantly(P<0.05).The apoptotic rate of cells transfected with si一SOX2OT increased significantly,and cell cycle was blocked at G0/G1 phase;si-SOX2OT inhibited cell migration and invasion.Conclusion The expression of LneRNA SOX2OT in EC is up-regulated.Inhibiting the expression of LneRNA SOX2OT can inhibit the proliferation of endothelial cells,block cell cycle,promote apoptosis and inhibit the invasion and migration of tumor cells.The study of SOX2OT provides an important theoretical basis for the diagnosis,treatment and prevention of metastasis and recurrenee of endothelial cells.
