摘要
目的以陕西不同产地太白楤木为原料,建立高效液相色谱法(HPLC)指纹图谱,并测定太白楤木中4种化学成分(楤木皂苷A,竹节参皂苷IVa,竹节参皂苷1,elatosideF)的含量。方法采用Thermo HyPURITY C18色谱柱(250 mm×4.6 mm,5μm),以乙腈(A)-0.2%磷酸水(B)溶液为流动相,梯度洗脱,柱温30℃,体积流量1 mL·min^-1,检测波长203 nm,进样量10μL。结果竹节参皂苷IVa,elatosideF,竹节参皂苷1,楤木皂苷A分别在0.5~14.4、0.4~41.3、10.8~223.8、27.5~275μg范围内线性关系良好;10批样品中上述4个成分的含量范围分别为0.219~2.021、0.184~3.434、4.398~89.230、14.714~47.902 mg·g^-1;建立了太白楤木的HPLC指纹图谱,得到16个共有峰,并识别了其中4个共有峰,相似度较好,聚类结果与相似度评价结果基本一致。结论该方法简单、灵敏、准确,可为太白楤木的鉴别及质量评价提供参考依据。
Objective To establish the HPLC fingerprints of Aralia taibaiensis from different habitats in Shaanxi Province and determine the contents of 4 chemical ingredients of araloside A,chikusetsusaponin IVa,chikusetsusaponin1 and elatosideF.Methods Thermo HyPURITY C18 column(250 mm×4.6 mm,5μm)was used for gradients elution with acetonitrile(A)-0.2%phosphoric acid(B)solution as mobile phase.Working conditions were as follows:the column temperature was 30℃,the flow rate was 1 mL·min^-1,the detection wavelength was 203 nm,and the injection volume was 10μL.Results The contents of chikusetsusaponin IVa,elatosideF,chikusetsusaponin 1 and araloside A had good linear relationships with the peak areas in the content ranges of 0.5-14.4,0.4-41.3,10.8-223.8,and 27.5-275μg,respectively.The contents range of the above 4 chemical ingredients in 10 batches of A.taibaiensis samples were 0.219-2.021,0.184-3.434,4.398-89.230,and 14.714-47.902 mg·g^-1,respectively.The HPLC fingerprint of A.taibaiensis was established and 16 common peaks were obtained,among which 4 peaks were identified.The overall similarity was good,and the clustering results were consistent with the similarity evaluation results on the whole.Conclusion This method was simple,sensitive and accurate,which provided a reference for the identification and quality evaluation of A.taibaiensis.
