摘要
目的:基于网络药理学方法探讨茯苓杏仁甘草汤与橘枳姜汤治疗冠状动脉粥样硬化性心脏病(简称冠心病)的作用靶点及可能作用机制。方法:利用TCMSP、DrugBank数据库,采用口服生物利用度和药物相似性筛选获取茯苓杏仁甘草汤与橘枳姜汤的成分和靶点,通过CTD、DrugBank数据库筛选抗冠心病相关的蛋白,构建成分-靶点网络图,用DAVID数据库对基因富集分析,通过KEGG通路分析结果,获得参与相关通路的候选基因及功能。结果:目前,茯苓杏仁甘草汤与橘枳姜汤的化学成分分别为186、163个;茯苓杏仁甘草汤与橘枳姜汤共有270个潜在靶点,其中256个潜在靶点与冠心病相关;成分-靶点网络图显示两方可能通过CHRM2、PTGS2、SLC6A2、CHRM1与PTGS1等关键基因,经由神经信号传递通路、cGMP-PKG信号通路、钙离子信号通道、血清素神经突触和甲状腺激素信号通路等干预冠心病的进程。茯苓杏仁甘草汤调节NOS2、PTGS2、F2、ABAT与PLA2G2E基因及Mapk信号通路和Egf信号通路等通路发挥特有的干预冠心病基础;F12、CHRM5、SELE、OPRD1与HAS2基因及抗生素的生物合成、脯氨酸代谢等通路的调控为橘枳姜汤特有的治疗冠心病机制。结论:茯苓杏仁甘草汤及橘枳姜汤药物之间存在相同及不同的化学成分及靶点效应干预冠心病,通过分析成分、靶点及通路的相互关系分析"同病异治"的科学内涵。
Objective: To explore the therapeutic target and possible mechanism of Fuling Xingren Gancao Decoction and Ju-ZhiJiang Decoction in treating coronary atherosclerotic heart disease( coronary heart disease) based on the method of network pharmacology. Methods: The component part and the therapeutic target of Fuling Xingren Gancao Decoction and Ju-Zhi-Jiang Decoction were selected with the TCMSP and Drug Bank database. Component-target network map was constructed by screening of proteins associated with coronary heart disease by CTD and Drug Bank Database,enrichment analysis of gene with DAVID database,candidate genes and functions involved in related pathways were obtained by results of KEGG pathway analysis. Results: Chemical constituents of Fuling Xingren Gancao Decoction and Ju-Zhi-Jiang Decoction were 186 and 163 respectively;there were 270 potential targets in Fuling Xingren Gancao Decoction and Ju-Zhi-Jiang decoction,which of 256 potential targets were associated with CHD;component-target network map demonstrates both decoctions probably with key gene of CHRM2,PTGS2,SLC6 A2,CHRM1,PTGS1 etc.,though intervention process of coronary heart disease of neural signal transduction pathway,c GMP-PKG signaling pathway,calcium signaling pathway,serotonin synapses and thyroid hormone signaling pathway. Fuling Xingren Gancao Decoction regulate NOS2,PTGS2,F2,ABAT and PLA2 G2 E gene and Mapk signaling pathway and Egf signaling pathway play a unique role in intervention of coronary heart disease;regulation of F12,CHRM5,SELE,OPRD1 and HAS2 genes,biosynthesis of antibiotics,proline metabolism and other pathways is the unique mechanism of Ju-Zhi-Jiang Decoction in the treatment of coronary heart disease. Conclusion: Between Fuling Xingren Gancao Decoction and Ju-Zhi-Jiang Decoction exist intervention of coronary heart disease with same and different chemical constituents and target effects. The scientific connotation of " treating same disease with different treatments " was analyzed by analyzing the relationship among components,targets and pathways.
