摘要
整体柱是一种用有机或无机聚合方法在色谱柱内进行原位聚合的连续床固定相。将温敏凝胶引入到整体柱体系中制备得到的温敏性整体柱,不仅可以使分离过程调控简单,还可以避免有机溶剂的使用,使整个分离过程绿色环保。文章综述了温敏-普通整体柱、温敏-毛细管柱、温敏-微流控柱、温敏-印迹柱和温敏-磁性柱的制备方法,以及近十五年来温敏性整体柱在核酸、类固醇、金属离子和其他物质分离、富集等方面中的应用。
Monolithic column is a continuous bed stationary phase which is polymerized in situ in chromatographic column by organic or inorganic polymerization method.The thermo-sensitive monolithic columns are obtained by introducing the thermo-sensitive hydrogels into the monolithic column system.It can quickly realize the efficient separation of small molecule drugs and biomacromolecules by adjusting the column temperature.It can not only make the control of separation process simple,but also avoid the use of organic solvents,making the whole separation process green and environmental protection.In this article,the preparation methods of the common thermo-sensitive monolithic column,the thermo-sensitive capillary column,the thermo-sensitive microfluidic column,the thermo-sensitive imprinted column and the thermo-sensitive magnetic column are reviewed.Furthermore,the applications of thermo-sensitive monolithic columns are introduced in aspects of separation and enrichment of nucleic acid,steroids,metal ions and other substances in recent 15 years.
