红土茯苓抑制5-LOX活性成分的筛选与分离 被引量:2

摘要 本实验以5-脂氧合酶为靶点,利用超滤液质联用技术筛选并鉴定红土茯苓中抑制5-脂氧合酶活性成分。结果表明,红土茯苓中白藜芦醇对5-脂氧合酶的抑制活性最强,其次为落新妇苷、黄杞苷、异黄杞苷和异落新妇苷。在此基础上首次采用高效逆流色谱技术,以正己烷∶正丁醇∶水(1∶2∶3∶v/v/v)为溶剂体系,从红土茯苓中分离得到落新妇苷、异落新妇苷、黄杞苷、异黄杞苷和白藜芦醇,经高效液相色谱分析,其纯度分别为98.39%,86.23%,97.15%,90.28%和99.29%。应用液-质联用技术,通过与标准化合物的保留时间、分子量,对分离得到五种化合物进行了结构鉴定。 Ultrafiltration LC-ESI-MSn was combined to screen 5-LOX inhibitors from Smilax china L for the first time.Results showed that,resveratrol was found to be strongest inhibtor towards 5-LOX,followed by astilbin,engelitin,isoengelitin,isoastilbin were also 5-LOX inhibitors in this assay.Five polyphenols,astilbin,isoastilbin,engelitin,isoengelitin and resveratrol were successfully isolated and purified from ethyl acetate extract of Smilax china L for the first time by high performance current-counter chromatography(HPCCC)using the solvent system Hex∶BuOH∶H2O at the volumn ratio 1∶2∶3,and purity of the five compounds determined by HPLC were 98.39%,86.23%,97.15%,90.28%and 99.29%,respectively.The structures of the five compounds were identified by congruent retention times,the data of the liquid chromatography-electrospray ionization-mass spectrometry(LC-ESI-MS)in the negative ion mode with those of the authentic standards and literature reports.
