HPLC测定莲不同部位紫云英苷及异鼠李素-3-O-葡萄糖苷含量

摘要 目的建立莲不同部位中紫云英苷及异鼠李素-3-O-葡萄糖苷含量的HPLC测定方法。方法采用Diamonsil C18色谱柱(200 mm×4.6 mm,5μm),以乙腈-0.5%甲酸溶液为流动相,梯度洗脱,检测波长347 nm,流速0.8 mL/min,柱温30℃,进样量10μL。结果在上述色谱条件下,紫云英苷、异鼠李素-3-O-葡萄糖苷与相邻色谱峰的分离度良好。紫云英苷在0.1005~2.01μg范围内有良好的线性关系(r=0.9996),平均加样回收率为100.96%(RSD=2.68%);异鼠李素-3-O-葡萄糖苷在0.026~0.52μg范围内有良好的线性关系(r=0.9994),平均加样回收率为98.91%(RSD=2.34%)。结论本研究建立的方法操作简便,结果可靠,可为莲相关药材的质量控制及资源开发提供依据。 Objective To establish an HPLC method for content determination of astragalin and isorhamnetin-3-O-glucoside in Nelumbo nucifera Gaertn.Methods The chromatographic separation was performed on a Diamonsil C18 column(200 mm×4.6 mm,5μm);the column temperature was maintained at 30℃;the mobile phase was acetonitrile-0.5%formic acid solution as gradient elution;the flow rate was 0.8 m L/min;the detection wavelength was at 347 nm;the injection volume was 10μL.Results Under above chromatographic conditions,astragalin and isorhamnetin-3-O-glucoside with adjacent other peaks were among the good separation.Astragalin had a good linear relationship in the range of 0.1005–2.01μg(r=0.9996);the average recovery was 100.96%(RSD=2.68%).Isorhamnetin-3-O-glucoside had a good linear relationship in the range of 0.026–0.52μg(r=0.9993);the average recovery was 98.57%(RSD=2.34%).Conclusion The method is convenient and reliable,which can provide evidence for the quality control and resource development of Nelumbo nucifera Gaertn.as a related medicine.
