干扰TRIM26表达对胶质瘤细胞增殖的影响

Effects of interfering TRIM26 expression on proliferation of glioma cells

目的:探讨干扰三结构域蛋白26(TRIM26)表达对胶质瘤细胞增殖的影响。方法:分别采用实时荧光定量PCR(qRT-PCR)和Western blot法检测正常胶质细胞HEB和胶质瘤细胞U251、U87、C6中TRIM26的表达水平。利用脂质体分别将TRIM26 siRNA(si-TRIM26组)或无义siRNA(si-NC组)转染到U251细胞中,将未转染的U251细胞记为空白对照组,转染48 h后分别采用qRT-PCR和Western blot法检测TRIM26 mRNA和蛋白的表达,转染24、48、72 h后用MTT法检测细胞增殖能力,转染48 h后用Western blot法检测细胞中PCNA和Ki-67的表达。结果:与HEB细胞相比,胶质瘤细胞中TRIM26的表达上调,U251细胞中的表达水平最高(P<0.05)。与空白对照组和si-NC组比较,si-TRIM26组U251细胞中TRIM26 mRNA和蛋白表达下降(P<0.05),细胞增殖能力下降(P<0.05),细胞中PCNA和Ki-67蛋白表达水平下降(P<0.05)。结论:干扰TRIM26的表达可抑制胶质瘤U251细胞的增殖。

Aim:To explore the effects of inhibiting the expression of tri-domain protein 26(TRIM26)by RNA interference on the proliferation of glioma cells.Methods:Real-time quantitative PCR(qRT-PCR)and Western blot were used to detect the expressions of TRIM26 mRNA and protein in normal glial HEB cells and glioma cells including U251,U87 and C6.U251 cells were transfected with TRIM26 siRNA(si-TRIM26 group)or the nonsense siRNA sequence(si-NC group)by liposome,and the untransfected U251 cells were the control group.After 48 hours transfection,the expressions of TRIM26 mRNA and protein were detected by qRT-PCR and Western blot,and PCNA and Ki-67 expression levels were detected by Western blot;the proliferation of cells was detected by MTT assay after 24,48,72 hours transfection.Results:Compared with normal glial HEB cells,the expressions of TRIM26 mRNA and protein in U87,C6,and U251 cells were significantly up-regulated,especially in U251 cells(P<0.05).Compared with control group and si-NC group,the expressions of TRIM26 mRNA and protein in si-TRIM26 group were significantly decreased(P<0.05),the proliferation of U251 cells was significantly lower(P<0.05),and the expressions of PCNA and Ki-67 protein were significantly lower(P<0.05).Conclusion:Interfering the expression of TRIM26 may inhibit the proliferation of glioma U251 cells.
