益气活血通阳法不同配比对急性心肌梗死兔血管生成因子影响的研究 被引量:2

Study on the effect of different ratios of Yiqi Huoxue Tongyang method on angiogenesis factors in rabbits with acute myocardial infarction

摘要 目的观察不同红参/水蛭配比组方的益气活血通阳之安心颗粒对急性心肌梗死兔梗死边缘区心肌及主动脉斑块内血管生成因子的影响,明确最佳配比组方。方法将50只健康雄性家兔按体质量随机分为10组,每组5只。假手术组给予正常饮食,只开胸穿线,不结扎左冠状动脉前降支,术后给予生理盐水灌胃。模型组、西药组和不同配比中药组复制动脉粥样硬化和急性心肌梗死双模型,模型组术后给予生理盐水灌胃,西药组于开胸结扎冠脉后当时立即在结扎处周围予碱性成纤维生长因子喷洒,不同配比中药组术后给予红参/水蛭比例为10/0,10/4,10/7,10/10,7/10,4/10,0/10的安心颗粒液连续灌胃10周。采用qPCR法检测各组家兔不同心肌区及主动脉区血管内皮生长因子受体-2(VEGFR-2)mRNA和低氧诱导因子-1α(HIF-1α)mRNA表达水平,采用Western Blot法检测各组家兔不同心肌区及主动脉区VEGFR-2和HIF-1α蛋白表达水平。结果模型组、西药组及不同配比中药组梗死心肌区和缺血心肌区VEGFR-2R、HIF-1αmRNA和蛋白表达水平均明显高于假手术组(P均<0.05)。西药组及10/4中药组缺血心肌区VEGFR-2R、HIF-1αmRNA和蛋白表达水平均显著高于其他中药组及模型组(P均<0.05),西药组与10/4中药组比较差异均无统计学意义(P均>0.05)。各组间主动脉区VEGFR-2R、HIF-1αmRNA和蛋白表达水平比较差异均无统计学意义(P均>0.05)。结论红参/水蛭比例为10/4的安心颗粒可提高梗死边缘区血管生成因子水平,且对动脉斑块内血管生成因子水平无明显影响。 Objective It is to observe the effects of Yiqi Huoxue Tongyang Anxin Granules with different red ginseng/leech combination prescription on angiogenesis factors in myocardial border zone and aortic plaques of rabbits with acute myocardial infarction,and to determine the optimal ratio.Methods Fifty healthy male rabbits were randomly divided into 10 groups according to their body weight,with 5 rats in each group.The sham operation group was given a normal diet,only the chest was threaded,the left anterior descending coronary artery was not ligated,and saline was administered intramuscularly.The model group,western medicine group and different proportions of traditional Chinese medicine group replicated the double model of atherosclerosis and acute myocardial infarction.The model group was given normal saline after operation,and the western medicine group was immediately given the alkali fibroblast growth factor by spraying around the ligation site after thoracotomy and ligation of the coronary artery.The different proportions of Chinese medicine groups were continuously treated with Anxin Granules at the proportions of 10/0,10/4,10/7,10/10,7/10,4/10,0/10 red ginseng/leech by gavage for 10 weeks.The expressions of vascular endothelial growth factor receptor-2(VEGFR-2)mRNA and hypoxia-inducible factor-1α(HIF-1α)mRNA in different myocardial and aortic regions of rabbits were detected by qPCR.The expression levels of VEGFR-2 and HIF-1αprotein in different myocardial and aortic regions of rabbits in each group was detected by Western Blot method.Results The expression levels of VEGFR-2R and HIF-1αmRNA and protein in the infarcted myocardium and ischemic myocardium of the model group,western medicine group and different proportions of Chinese medicine group were significantly higher than those of the sham operation group(P<0.05).The expression levels of VEGFR-2R and HIF-1αmRNA and protein in the ischemic myocardium of western medicine group and 10/4 Chinese medicine group were significantly higher than those of other traditional Chinese medicine group and model group(P<0.05),the differences were not statistically significant between the western medicine group and 10/4 Chinese medicine group(P>0.05).There were no significant differences in the expression levels of VEGFR-2R and HIF-1αmRNA and protein in the aortic area between these groups(P>0.05).Conclusion Anxin granules with a ratio of 10/4 red ginseng/leech can increase the level of angiogenic factors in the infarct border zone and have no significant effect on the level of angiogenic factors in arterial plaque.
