摘要
目的:研究内胚层分化过程中初级纤毛的形态变化及纤毛合成有关基因的表达变化。方法:通过免疫荧光检测人胚胎干细胞和内胚层细胞内中心粒周蛋白及乙酰化微管蛋白的表达,利用qRT-PCR测定纤毛合成有关基因的表达。结果:初级纤毛同时存在于两种细胞中;与人胚胎干细胞比较,内胚层细胞的纤毛明显缩短,KIF17和IFT172基因表达显著下调。结论:初级纤毛在内胚层分化过程中会变短。
Objective: To investigate the morphologic changes of primary cilia and the expression of ciliogenesis-related genes during the endodermal differentiation from human embryonic stem cells(hESCs). Methods: Pericentrin and acetylated-tubulin in both hESCs and definitive endoderm(DE) were detected by immunofluorescence, and the expression of genes related to cilia synthesis was determined by qRT-PCR. Results: Primary cilia in DEs were shorter than those in hESCs, and gene expression of KIF17 and IFT172 was significantly reduced. Conclusions: Primary cilia shortened largely in endodermal differentiation.
