Cofilin-1与肝癌细胞迁移侵袭能力的相关性研究 被引量:7

摘要 探讨丝切蛋白1(Cofilin-1)与肝癌细胞的迁移、侵袭能力的关系,运用免疫印迹试验(western blot)检测肝癌细胞(Huh-7细胞)、转染上皮生长因子受体的人肝癌细胞(Huh-7-EGFR细胞)与转染上皮生长因子受体突变体Ⅲ的人肝癌细胞(Huh-7-EGFRvⅢ细胞)三种细胞中Cofilin-1的蛋白表达。然后将Cofilin-1用慢病毒表达系统导入Huh-7细胞,构建转染丝切蛋白1的人肝癌细胞(Huh-7-Cofilin-1细胞)系,并应用迁移侵袭实验检测细胞迁移侵袭的能力;再用RNA干扰(siRNA)技术沉默Huh-7-EGFRvⅢ细胞中的Cofilin-1,然后应用迁移侵袭实验检测细胞的迁移侵袭能力。结果显示:转染Cofilin-1后,Huh-7细胞的迁移侵袭能力显著增强(P<0.05);而干扰Cofilin-1后,Huh-7-EGFRvⅢ细胞迁移与侵袭能力明显减弱(P<0.05)。表明Cofilin-1参与肝癌细胞迁移与侵袭。 In order to study the relationship between cofilin-1 and the migration and invasion ability of hepatocellular carcinoma cells.The protein expression of Cofilin-1 in Huh-7 cells and Huh-7-EGFR cells and Huh-7-EGFRv Ⅲ cells were detected by western blot.Cofilin-1 was transfected into Huh-7 cells by lentivirus expression system.Cofilin-1 was silenced in the Huh-7-EGFRv Ⅲ cells.The migration or invasion ability of cells were analyzed and verified by transwell migration assay and matrigel invasion assay.The protein expression of Cofilin-1 in Huh-7-EGFRv Ⅲ cells was significantly higher than that in Huh-7 and Huh-7-EGFR cells.After transfection of Cofilin-1,the migration and invasion ability of Huh-7 cells were enhanced.Conversely,after interference Cofilin-1,the migration and invasion ability of Huh-7-EGFRv Ⅲ cells were decreased.In conclusion,the migration and invasion of Hepatocellular Carcinoma cells were promoted by Cofilin-1.
