经血来源间充质干细胞对大鼠薄型子宫内膜的治疗效果 被引量:5

摘要 目的探究经血来源间充质干细胞(menstrual blood-derived stem cells,MenSCs)对大鼠薄型子宫内膜的治疗效果。方法将20只无特殊病原体(specific pathogen free,SPF)雌性大鼠随机分为对照组、模型组、MenSCs组和雌激素组。模型组、MenSCs组、雌激素组大鼠采用子宫注射无水乙醇的方法构建薄型子宫大鼠模型,MenSCs组大鼠尾静脉注射MenSCs,雌激素组大鼠尾静脉注射雌激素。于3个动情周期后处死大鼠,取其子宫内膜标本制作切片,比较四组大鼠子宫内膜厚度和角蛋白、波形蛋白、整合素β3的表达情况。结果分离培养的MenSCs表现出明显的间充质干细胞特征,高表达CD44、CD73、CD90、CD105和OCT-4、C-kit、波形蛋白,不表达HLA-DR。成骨、成脂、成软骨分化均为阳性。模型组、MenSCs组、雌激素组大鼠子宫内膜厚度均显著小于对照组(均P<0.001),其中模型组大鼠子宫内膜厚度最小,MenSCs组大鼠子宫内膜厚度显著大于雌激素组(P=0.047)。模型组、MenSCs组、雌激素组大鼠子宫内膜组织角蛋白、波形蛋白、整合素β3表达水平均显著低于对照组(均P<0.05),MenSCs组大鼠上述蛋白表达水平均显著高于模型组和雌激素组(均P<0.05)。结论MenSCs可改善大鼠薄型子宫内膜的容受性和功能,其治疗效果优于雌激素治疗。 Objective To explore curative effect of menstrual blood-derived stem cells(MenSCs)on thin endometrium in rats.Method 20 female specific pathogen free(SPF)rats were randomly divided into control group,model group,MenSCs and estrogen group.In model group,MenSCs group and estrogen group,uterine injection of anhydrous ethanol was performed to construct thin uterus rat models.MenSCs group and estrogen group rats were given tail vein injection of MenSCs and estrogen,respectively.The rats were sacrificed after 3 estrus cycles,and endometrial specimens were collected.The endometrial thickness,the expression of keratin,vimentin and integrinβ3 were compared among the four groupps.Result The separately cultured MenSCs cells showed obvious features of mesenchymal stem cell,high expression of CD44,CD73,CD90,CD105 and OCT-4,C-kit,vimentin,nonexpression of HLA-DR.The differentiation of osteogenesis,lipogenesis and chondrogenesis were positive.Endometrial thickness in model group,MenSCs group and estrogen group was significantly smaller than that of control group respectively(all P<0.001),which was the smallest in model group,the endometrial thickness in MenSCs group was larger than that in estrogen group(P=0.047).The expression level of keratin,vimentin and integrinβ3 in endometrium tissue of model group,MenSCs group and estrogen group were lower than those of control group(all P<0.05).The expression level of above protein in MenSCs group were significantly higher than those in model group and estrogen group(all P<0.05).Conclusion MenSCs can improve receptivity and function of thin endometrium,whose curative effect is better than that of estrogen alone.
