摘要
目的通过检测喉癌细胞BNIP3基因表达的阳性率,探讨BNIP3基因在喉癌细胞中的表达情况。通过检测不同浸润深度、不同淋巴结转移情况、不同分期、不同分化程度的喉癌细胞中BNIP3基因表达的阳性率,探讨BNIP3基因在喉癌中表达的影响因素。方法2005年1月~2018年12月收入我院头颈外科治疗的270例喉癌患者,均经临床和病理明确诊断,取其肿瘤组织细胞检测,设为喉癌组;另取对应粘膜组织为对照组。应用组织芯片免疫组化S-P染色技术测定各组细胞中BNIP3基因表达情况。结果①喉癌组BNIP3基因表达阳性率低于对照组,差异有统计学意义(P<0.05)。②与浅度浸润组比较,深度浸润组BNIP3基因表达阳性率明显降低,差异有统计学意义(P<0.05);与对照组比较,浅度浸润组和深度浸润组BNIP3基因表达阳性率有不同程度降低,差异有统计学意义(P<0.05)。③与无淋巴结转移组比较,有淋巴结转移组BNIP3基因表达阳性率明显降低,差异有统计学意义(P<0.05);与对照组比较,无淋巴结转移组及有淋巴结转移组BNIP3基因表达阳性率有不同程度降低,差异有统计学意义(P<0.05)。④与早期喉癌组比较,中晚期喉癌组BNIP3基因表达阳性率明显降低,差异有统计学意义(P<0.05);与对照组比较,早期喉癌组和中晚期喉癌组BNIP3基因表达阳性率有不同程度降低,差异有统计学意义(P<0.05)。⑤与高分化组比较,中分化组和低分化组BNIP3基因表达阳性率均有不同程度降低,差异有统计学意义(P<0.05);与中分化组比较,低分化组BNIP3基因表达阳性率显著降低,差异有统计学意义(P<0.05);与对照组比较,高分化组、中分化组及低分化组BNIP3基因表达阳性率均有不同程度降低,差异有统计学意义(P<0.05)。⑥胞浆内的特异性染色(BNIP3)在周边区域的癌细胞中表现为深染,比肿瘤中心区域和正常粘膜区域的染色深。结论喉癌细胞中BNIP3基因表达与浸润深度、淋巴结转移情况、TNM分期、肿瘤细胞分化程度有一定关系。
Objective To investigate the expression of BNIP3 gene in laryngocarcinoma by detecting the positive ratio of BNIP3 gene expression in different depth of invasion,different condition of lymph node metastasis,different stages,different differentiation degrees of laryngocarcinoma.Methods From Jan 2005 to Dec 2018,270 cases of laryngocarcinoma patients treated in our hospital were set as laryngocarcinoma group.270 cases of corresponding normal tissue mucosa samples were set as control group.Applicated immunohistochemistry S-P dyeing technology to determinate expression of BNIP3 gene expression in each group.Results①The positive ratio of BNIP3 gene in laryngocarcinoma group was significantly lower than that in control group,and the difference had statistical significance(P<0.05).②Compared with shallow invasion group,deep invasion group of positive rate of BNIP3 gene expression significantly reduced,and the difference had statistical significance(P<0.05).Compared with control group,shallow invasion group and deep invasion group of positive rate of BNIP3 gene expression had different degrees of reduction,and the difference had statistical significance(P<0.05).③Compared with no lymph nodes metastasis group,lymph nodes metastasis group of positive rate of BNIP3 gene expression significantly reduced,and the difference had statistical significance(P<0.05).Compared with control group,no lymph nodes metastasis group and lymph nodes metastasis group of positive rate of BNIP3 gene expression had different degrees of reduction,and the difference had statistical significance(P<0.05).④Compared with early laryngocarcinoma group,advanced laryngocarcinoma group of positive rate of BNIP3 gene expression significantly reduced,and the difference had statistical significance(P<0.05).Compared with control group,early laryngocarcinoma group and advanced laryngocarcinoma group of positive rate of BNIP3 gene expression had different degrees of reduction,and the difference had statistical significance(P<0.05).⑤Compared with high differentiation group,intermediate and low differentiation group of positive rate of BNIP3 gene expression had different degrees of reduction,and the difference had statistical significance(P<0.05).Compared with intermediate differentiation group,low differentiation group of positive rate of BNIP3 gene expression significantly reduced,and the difference had statistical significance(P<0.05).Compared with control group,high,intermediate and low differentiation group of positive rate of BNIP3 gene expression had different degrees of reduction,and the difference had statistical significance(P<0.05).⑥In laryngocarcinoma surrounding areas,specific staining in cytoplasm(BNIP3)showed dark stained,deeper than centre areas of laryngocarcinoma and normal mucosa.Conclusion The expression of BNIP3 gene is partly correlated with the depth of invasion,TNM stage,lymph node metastasis and differentiation.
