Digital PCR Technology and Its Applications in Forensic Science

摘要 数字PCR(dPCR)是一种核酸定量分析的新兴技术手段,可以直接计数DNA分子个数,对起始样品进行绝对定量。与传统PCR技术相比,dPCR不需要建立标准曲线,具有良好的应用前景。同时dPCR技术在法庭科学领域具有独特而广阔的应用前景。通过阐述dPCR技术的发展、原理、特点及其在法庭科学中的下一代测序、种属鉴定和定量、病原微生物检测、甲基化水平检测等方面的应用,总结该项技术所面临的挑战,同时对其应用前景进行展望。 Digital PCR(dPCR)is an emerging technology for quantitative analysis of nucleic acids.Digital PCR can directly count the number of DNA molecules and perform absolute quantification on the initial samples.Compared with traditional PCR technology,dPCR does not need to establish a standard curve,so it has good application prospects.At the same time,dPCR technology has unique and broad application prospects in the field of forensic science.In this paper,the developments,principles and characteristics of dPCR technology and its applications in forensic science,such as next generation sequencing,species identification and quantification,pathogenic microorganism detection,methylation level detection,etc.are described.The challenges faced by dPCR technology are summarized,and its application prospects are prospected.
