电针对脑卒中痉挛状态大鼠黑质内多巴胺受体及其亚型含量与表达的影响 被引量:8

摘要 目的通过动物实验分析电针治疗对脑卒中痉挛状态下SD大鼠大脑黑质中多巴胺受体及其亚型的含量与表达的影响,探讨电针缓解脑卒中痉挛状态(SCA)的疗效及其作用机制。方法将32只SD大鼠随机分为电针组(模型+电针阳陵泉、曲池)、假手术组、模型组、空白组,每组8只。运用在大脑中动脉线栓法制作SCA模型,行为学评分确认模型成功后开始电针治疗,连续3 d。酶联免疫吸附实验法(ELISA)检测黑质中多巴胺(DA)含量,以及DA的D1与D2受体的信使核糖核酸的含量,并通过逆转录-聚合酶链反应(RT-PCR)检测黑质中D1、D2两个亚型(DRD1 mRNA、DRD2 mRNA)的表达。结果(1)神经功能损伤评分在电针治疗前,模型组、电针组与空白组、假手术组比较评分升高(P<0.01);电针组治疗前后差异明显(P<0.01)。(2)模型组与空白组、假手术组比较,DA、DRD1、DRD2含量明显降低(P<0.01),电针组与模型组比较,DA、DRD1、DRD2含量明显升高(P<0.01)。(3)模型组与空白组、假手术组比较,DRD1 mRNA、DRD2 mRNA表达明显降低(P<0.01);电针组与模型组比较,表达均有增加(P<0.01)。结论电针治疗可调节大脑黑质内DA及其受体的含量与表达,从而改善中枢神经系统的功能。 Objective To observe the experimental effect of electroacupuncture on the content and expression of DA receptor and subtypes in substantia nigra of SD rats with spasm of cerebral apoplexy(SCA),and to investigate the effect and mechanism of electroacupuncture in relieving SCA.Methods 32 SD rats were randomly divided into the electroacupuncture group(model+electroacupuncture on GB 34 and LI 11),sham-operation group,model group,blank group,with 8 rats in each group.SCA model was built by middle cerebral artery occlusion and electroacupuncture was performed for 3 days after the model was successful built confirmed by the behavior score.Enzyme-linked immunosorbent assay(ELISA)was applied to detect the content of DA in substantia nigra and mRNA of D1 and D2 receptors.By RT-PCR,expressions of D1 and D2 subtypes(DRD1 mRNA,DRD2 mRNA)were detected.Results(1)neurological severity score(NSS)in the model group and the electroacupuncture group were higher than the blank group and sham-operation group before electroacupuncture treatment(P<0.01)and the difference was significant in the electroacupuncture group before and after treatment(P<0.01).(2)DA,DRD1,DRD2 in the model group decreased significantly compared with those in the blank group and the sham-operation group(P<0.01).DA,DRD1,DRD2 in the electroacupuncture group increased significantly,compared with the model group(P<0.01).(3)DRD1 mRNA,DRD2 mRNA in the model group decreased significantly compared with those in the blank group and sham-operation group(P<0.01);compared with those in the model group,the expressions in the electroacupuncture group increased significantly(P<0.01).Conclusion Electroacupuncture can regulate the content and expression of DA and its receptors in substantia nigra,to improve the function of central nervous system,showing that the electroacupuncture has a good treatment effect on SCA.
