摘要
目的:探讨附子理中汤治疗胃溃疡的药效机制。方法:用食醋连续灌胃建立大鼠胃溃疡模型,实验设空白组、模型组、西药雷尼替丁组、附子理中汤组。灌胃给予相应的临床等剂量药物7天后,处理动物,进行指标检测。①常规方法检测大鼠体质量、饮食、胃指数和脾指数;②HE染色观察大鼠胃肠组织病理;③RT-PCR法检测大鼠胃组织中AQP1、AQP3和AQP4mRNA表达情况。结果:①附子理中汤能有效改善胃溃疡模型大鼠竖毛、弓背、蜷卧、便溏、饮食下降、消瘦等情况;②附子理中汤能有效改善胃溃疡模型所致的胃黏膜破坏、上皮脱落,炎细胞浸润,炎性物质渗出及水肿等情况;③附子理中汤能有效提高AQP1mRNA的表达,下调胃溃疡模型大鼠胃组织中AQP3和AQP4mRNA的表达水平。结论:附子理中汤可能是通过调节水液代谢平衡,促进AQP1,抑制AQP3、AQP4的表达,从而达到改善大鼠胃组织溃疡和水肿程度。
Objective:To study the pharmacodynamic mechanism of Fuzi Lizhong Decoction in the treatment of gastric ulcer.Methods:The model of gastric ulcer was established by continuous gavage with vinegar in the rats.The model rats were randomly divided into the model group,the western medication group(Ranitidine),Fuzi Lizhong Decoction group,the normal rats were taken as the blank control.After 7 days of corresponding intervention,body weight,diet condition,gastric index and spleen index were examined,pathological changes of gastrointestinal tissues were observed by HE staining method,the mRNA expressions of AQP1,AQP3 and AQP4 in stomach were detected by RT-PCR technique.Results:Fuzi Lizhong Decoction can effectively improve the rat model of gastric ulcer,such as vertical hair,bow-back,curling,loose stools,poor diet and emaciation,it can effectively improve gastric mucosal destruction,epithelial exfoliation,inflammatory cell infiltration,inflammatory substance exudation and edema caused by gastric ulcer,it also can effectively up-regulate the expression of AQP1 mRNA and down-regulate the expressions of AQP3 mRNA and AQP4 mRNA in gastric tissues.Conclusion:Fuzi Lizhong Decoction can improve the degree of gastric ulcer and edema of the rats with gastric ulcer by regulating water and liquid metabolism,promoting the expression of AQP1 mRNA and inhibiting the expressions of AQP3 mRNA and AQP4 mRNA.
