肝纤维化形成过程中血清ROS和GSH的水平变化 被引量:5

摘要 目的研究肝纤维化形成过程中血清活性氧簇(ROS)和还原性谷胱甘肽(GSH)的水平变化及其相互关系,探讨其在肝纤维化形成中的意义。方法 34只健康雄性SD大鼠随机分两组:对照组6只和模型组28只。模型组给予皮下注射40%CCl4油剂,对照组给予相同剂量的油剂皮下注射,注射CCl4后的第2,4,6,8周各杀死模型组大鼠6只,对照组在8周时杀死6只大鼠,留取血清及肝组织待测。收集正常健康对照组及乙肝肝硬化组血清标本各20例,采用比色法分别测定各组血清中ROS及GSH的浓度。结果模型组2,4,6,8周大鼠的ROS水平均较对照组大鼠升高(P <0. 05);模型组2,4,6,8周大鼠的GSH水平与对照组比较,差异无统计学意义(P> 0. 05)。肝硬化患者血清ROS浓度明显高于正常组(P <0. 05),而血清GSH浓度与正常组比较,差异无统计学意义(P> 0. 05)。结论肝纤维化形成过程中ROS、GSH的表达水平升高,提示可能参与了肝纤维化的发生。 Objective To investigate the changes of reactive oxygen species( ROS) and reduced glutathione( GSH) and explore their correlation and clinical significance in the development of hepatic fibrosis. Methods Thirty-four healthy male SD rats were randomly assigned to control group( n = 6) and model group( n = 28). Rats were subcutaneously injected with 40% CCl4 oil solution in model group,and the same volume of oil solution in control group. Six rats were killed at 2,4,6,8 week after CCl4 treatment,respectively.Six rats in control group were sacrificed at 8 week to take serum and liver tissues for testing. Serum samples from 20 cases of normal healthy controls and 20 cases of HBV-related cirrhosis were collected. ROS and GSH concentrations in serum were detected in each group using the colorimetric method. Results The level of ROS in the rats was higher in model group at 2,4,6 and 8 week than in control group( P < 0. 05). The level of GSH showed no significant difference between model group at 2,4,6 and 8 week and control group( P > 0. 05). Serum ROS was significantly higher in patients with HBV-related cirrhosis than in healthy controls( P < 0. 05),and serum GSH showed no significant difference between cirrhosis patients and normal controls( P > 0. 05). Conclusion The expression of ROS,GSH in the process of hepatic fibrosis is increased,indicating that they may play a role in the development of hepatic fibrosis.
