As a member of the late expression factors(LEFs)family,LEF-6 of Bombyx mori nucleopolyhedrovirus(BmNPV)is able to accelerate late gene transcription.In order to study the function of lef-6 gene in the process of viral infection and influence on host cell,we constructed a lef-6-null Bacmid(lef-6-ko-Bacmid)by deleting lef-6 gene with Red recombinant technology,transfected BmN cell,and then ectopia repaired lef-6 by Bac-to-Bac system to constructe a lef-6 repaired Bacmid(lef-6-re-Bacmid).qRT-PCR results showed that deletion of lef-6 gene resulted in decrease in viral genome replication,as well as decline in transcription levels of genes at different stages(early,late and very late).In addition,transcription level of apoptosis suppressor gene iap2 also decreased.Further Western blot analysis showed that overexpression of lef-6 could elevate the expression level of Bcl-2 protein and reduce the expression level of Bax protein to prevent cell apoptosis.In conclusion,deletion of lef-6 gene not only leads to a drop in viral genome replication and gene transcription,but also affects the process of host cell apoptosis by regulating the expression level of Bcl-2 and Bax protein.These results provided new references for exploring the interactive mechanism between BmNPV and silkworm.
