摘要
目的:探讨不同浓度孔雀草花水提物对四氯化碳(CCl4)致小鼠急性肝损伤小鼠的保护作用。方法:将50只小鼠随机分成5组,每组10只,分别为空白对照组,模型组,孔雀草低、中、高剂量组(1.126 g/kg、2.252 g/kg、4.504 g/kg)。各给药组均给予相应药物,空白组与模型组给予同体积生理盐水,连续灌胃10 d。末次灌胃2 h后,除空白组腹腔注射植物油溶液外,其余各组均腹腔注射0.5%四氯化碳植物油溶液,造模剂量为10 mL/kg,染毒24 h建立急性肝损伤模型,测定并比较各组小鼠血清谷丙转氨酶(ALT)和谷草转氨酶(AST)活性、肝组织超氧化物歧化酶(SOD)活力、丙二醛(MDA)含量。采用酶联免疫法测定肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)并观察肝脏病理学变化的差异。结果:低、中、高剂量孔雀草花水提物均能明显降低CCl4致肝损伤小鼠ALT、AST活性,升高肝组织中SOD活力,降低MDA含量,且能够有效改善炎症因子TNF-α和IL-6的表达,与模型组比较差异具有统计学意义(P<0.01)。镜下观察表明,与模型组相比,不同浓度的孔雀草花水提物均可缓解肝受损情况。结论:孔雀草花水提物对四氯化碳致急性肝损伤的小鼠具有保护作用,其作用机制可能与抗氧化、抗炎有关。
Objective:To explore the protective effects of different concentrations of water extract of Tagetes patula L flower on carbon tetrachloride(CCl4)-induced acute hepatic injury in mice.Methods:50 mice were randomly divided into 5 groups,10 in each group,which were blank control group,model group and low-dose,mid-dose and high-dose groups of Tagetes patula L(1.126 g/kg、2.252 g/kg、4.504 g/kg)flower.Each drug group is given the corresponding drug,The blank group and the model group were given equal volume of saline by gavage,all groups were lavaged for 10 d.Two hours after the last gavage,except for the intraperitoneal injection of vegetable oil solution in the blank group,the other groups were given intraperitoneally injected with 0.5%carbon tetrachloride vegetable oil solution at a dose of 10 mL/kg,and an acute liver injury model was established after 24 hours of exposure.The serum alanine aminotransferase(ALT)and aspartate aminotransferase(AST),the content or activity of liver malondialdehyde(MDA),superoxide dismutase(SOD)were measured and compared.Tumor Necrosis Factor-α(TNF-α)、Interleukin-6(IL10)expression levels were measured by enzymelinked immunosorbent assay(ELISA)and observation of differences in liver pathology.Results:The activities of ALT,AST,TNF-αand IL10 as well as level of hepatic MDA were significantly decreased by low,medium and high dose of extract of Tagetes patula L while activity of SOD in liver tissue was markedly increased.Compared with the model group,the difference is significant and highly statistically significant(P<0.01).Microscopic observation of different concentrations of aqueous extracts compared with the model group can alleviate liver damage.Conclusion:The water extracts of Tagetes patula L have protective effect on acute hepatic injury induced by CCl4 in mice.Its mechanism may be related to anti-oxidative and anti-inflammatory.
