利用MAS进行大豆花叶病毒SC7抗性鉴定及分子育种初探 被引量:6

摘要 大豆花叶病毒株系SC7是黄淮海和长江流域大豆产区主要的一种流行株系,基于对传统育种与分子育种相结合来选育抗病品种需求,为发掘一种辅助鉴定大豆抗花叶病毒SC7的引物及其检测方法应用于一线育种。由大豆公共遗传图谱上挑选的引物筛选比对24个自然群体材料,确定其引物BARCSOYSSR_02_0631抗病(包括中感)检出符合率为64.3%,感病符合率为100%,即检测正确率为79.2%,而且经过室内辅助选择育种鉴定到了一批能进入安徽省区域试验和生产试验的抗花叶病毒病SC7的新品系阜Y31和阜豆169等,通过该标记对大豆品系抗花叶病毒SC7的相关位点进行早期子代选择鉴定,该辅助检测方法不仅绕开了试验站实验条件的限制而且避免了传统方法接种鉴定花叶病毒的传播,减少了随后的育种工作量和育种周期,利用MAS技术可快速筛选出抗病高产的大豆育种材料。 Soybean mosaic virus(SMV)strain SC7 is a major epidemic strain to the soybean production in Huang-Huai-Hai area and Yangtze River valley.Based on the demand of combining traditional breeding with molecular breeding to select disease-resistant varieties,a primer for assisting identification of soybean mosaic virus SC7 and its detection method were developed and applied to the first-line breeding.By comparing 24 natural population materials selected from soybean public genetic map of markers,the coincidence rate of BARCSOYSSR_02_0631 primer for disease resistance(including moderate susceptibility)was 64.3%,and the coincidence rate of susceptibility was 100%.That is,the correct rate of detection was 79.2%.Moreover,FuY31 and Fudou 169,a series of lines were identified and selected to the regional trial and production trial in Anhui Province.The marker was used to select and identify the related loci of SC7 resistant to soybean mosaic virus in the early generation.The assistant detection method not only circumvented the limitation of experimental conditions in the test station,but also avoided the transmission of traditional methods to inoculate and identify mosaic virus.By using MAS technology,soybean breeding lines with high yield and disease resistance could be quickly screened out.
