妊娠期细颗粒物暴露致小鼠子代海马神经炎症发生

Exposure to fine particles during pregnancy causes neuroinflammation in the hippocampus of offspring mice

摘要 目的观察妊娠期PM2.5暴露后出生后21d子代鼠海马炎症因子的表达变化。方法孕鼠随机分为空白组、对照组、PM2.5低剂量组、PM2.5中剂量组、PM2.5高剂量组,利用气管滴注法分别向PM2.5低、中、高剂量组滴注不同浓度的PM2.5悬浊液,对照组滴注相同体积PBS,空白组不做处理。孕期间断染毒6次。实时荧光定量PCR技术检测出生后21d子代鼠海马中肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)和干扰素-γ(IFN-γ)mRNA含量变化情况,酶联免疫技术检测炎症因子TNF-α、IL-1β和IFN-γ蛋白表达情况。结果与对照组相比,PM2.5高剂量组子代鼠海马中IL-1βmRNA含量升高,PM2.5中、高剂量组TNF-α和IFN-γmRNA含量均显著升高;PM2.5模型组海马TNF-α、IL-1β和IFN-γ蛋白表达量均明显升高。结论妊娠期PM2.5暴露可致子代鼠出生后海马炎症因子TNF-α、IL-1β和IFN-γ含量改变,其改变可能会导致子代神经炎症发生。 Objective The expression changes of inflammatory factors in hippocampus of offspring rats were studied 21 days after birth after PM2.5 exposure.Methods Pregnant mice were divided into blank group,control group,PM2.5 low-dose group,PM2.5 medium-dose group,and PM2.5 high-dose group randomly.The different concentrations of PM2.5 suspension were given to low,medium and high-doses group of PM2.5 by tracheal instillation.The control group was instilled with the same volume of PBS,and the blank group was not.Six times of infection during pregnancy.The newborn mice were sacrificed to take hippocampus after 21 days of birth.Then the real-time quantitative PCR was used to detect the changes of Tumor necrosis factor-α(TNF-α),Interleukin-1β(IL-1β) and Interferon-γ(IFN-γ) mRNA in hippocampus of newborn mice.And the expression of inflammatory factors TNF-α,IL-1β and IFN-γ protein were detected by enzyme-linked immunosorbent assay.Results Compared with the control group,the IL-1β mRNA content in the hippocampus of the PM2.5 high-dose group was increased,and the TNF-α and IFN-γ mRNA levels were significantly increased in PM2.5 medium-dose group and high-dose group;the expressions of TNF-α,IL-1β and IFN-γ protein in hippocampus of PM2.5 dose group were significantly increased.Conclusion Exposure to PM2.5 during pregnancy can cause changes in the levels of inflammatory factors TNF-α,IL-1β and IFN-γ in the hippocampus after birth,which may lead to neuroinflammation in offspring.
