小檗碱对多囊卵巢综合征大鼠激素水平及胰岛素抵抗的影响及机制 被引量:7

Effect and mechanism of berberine on hormone levels and insulin resistance of polycystic ovary syndrome rats

摘要 目的研究小檗碱对多囊卵巢综合征大鼠激素水平及胰岛素抵抗的影响,并初步分析其作用机制。方法将45只雌性SD大鼠随机分为对照组、模型组及实验组,各15只。模型组及实验组均连续20 d皮下注射脱氢表雄酮构建多囊卵巢综合征大鼠模型,建模成功后实验组灌胃1.5 g·kg^-1小檗碱,对照组与模型组灌胃等量生理盐水,连续干预2周。以化学免疫发光法检测血清激素水平,以血糖仪检测血糖、胰岛素水平,计算胰岛素抵抗指数(HOMA-IR);以蛋白质印迹(WB)法检测卵巢组织PI3K/Akt信号通路相关蛋白表达。结果对照组、模型组及实验组大鼠血清睾酮(T)水平分别为(4.12±1.56),(15.02±1.89),(7.11±1.25)ng·m L^-1,促黄体激素(LH)水平分别为(4.12±0.99),(6.33±1.02),(5.01±0.84)m U·m L^-1,及促卵泡素(FSH)水平分别为(2.85±0.51),(1.61±0.37),(2.26±0.41)m U·m L^-1,空腹血糖(FBG)水平分别为(4.33±0.98),(6.12±1.03),(5.09±0.87)mmol·L^-1,空腹胰岛素(FINS)水平分别为(4.01±0.85),(6.74±0.96),(5.21±0.79)m U·L^-1,HOMA-IR分别为0.76±0.46,1.89±0.55,0.85±0.47,卵巢组织磷脂酰肌醇3-激酶(PI3K)蛋白相对表达量分别为0.89±0.10,0.45±0.12,0.76±0.09,蛋白激酶B(Akt)蛋白相对表达量分别为0.72±0.08,0.61±0.14,0.82±0.09,差异均有统计学意义(均P<0.05)。结论小檗碱可有效调节多囊卵巢综合征大鼠模型血清激素水平,降低胰岛素抵抗,其作用机制与有效活化卵巢组织PI3K/Akt信号通路相关。 Objective To explore the effect of berberine on hormone levels and insulin resistance of polycystic ovary syndrome rats,analyse the action mechanism.Methods Forty-five female rats were randomly divided into control group,model group and test group,15 rats in each group.Model group and test group were built polycystic ovary syndrome rats model by subcutaneous injection dehydroepiandrosterone for 20 d continuously,test group was treated with gastrointestinal administration of1.5 g·kg^-1 berberine after the success of the modeling,control group and model group were treated with gastrointestinal administration the same amount of normal saline,intervened for 2 weeks continuously.The serum hormone levels was detected by chemical immune luminescence method,the blood glucose and insulin levels were detected by glucosemeter,homeostasis model assessment for insulin resistance(HOMA-IR)was calculated;the PI3 K/Akt signaling pathways related protein expression in ovarian tissue was detected by Western blot(WB)method.Results The serum testosterone(T)levels in control group,model group and test group were(4.12±1.56),(15.02±1.89),(7.11±1.25)ng·m L^-1,luteinizing hormone(LH)levels were(4.12±0.99),(6.33±1.02),(5.01±0.84)m U·m L^-1,follicle stimulating hormone(FSH)levels were(2.85±0.51),(1.61±0.37),(2.26±0.41)m U·m L^-1,fasting blood glucose(FBG)levels were(4.33±0.98),(6.12±1.03),(5.09±0.87)mmol·L^-1,fasting insulin(FINS)levels were(4.01±0.85),(6.74±0.96),(5.21±0.79)m U·L^-1,HOMA-IR were 0.76±0.46,1.89±0.55,0.85±0.47,ovarian tissue phosphatidyl inositol 3 kinase(PI3 K)relative expression were 0.89±0.10,0.45±0.12,0.76±0.09,Protein kinase B(Akt)relative expression were 0.72±0.08,0.61±0.14,0.82±0.09,all with significant difference(all P<0.05).Conclusion Berberine can adjust the serum hormone levels of polycystic ovary syndrome rats,reduce the insulin resistance,the action mechanism is related to the effective activation of ovarian tissue PI3 K/Akt signaling pathway.
