AILTV临床分离株鸡肝癌细胞培养特性及黏附因子测定

The influence of AILTV on growth characteristics and adhesion factor in chicken liver cancer cells

摘要 禽传染性喉气管炎病毒(avian infectious laryngotracheitis virus,AILTV)引起鸡上部呼吸道传染病,危害养鸡业的发展。2017年冬季至2018春季,本实验室从河北地区各发病鸡场病死鸡分离鉴定得到8株AILTV,分别编号为AILTV-HB-1~AILTV-HB-8。将病毒进行鸡胚传代后,与标准强毒王岗株和河北地区使用的2株疫苗分别接种于鸡肝癌细胞(LMH),检测LMH增殖速度。ELISA方法检测鸡整合素和鸡层黏连蛋白/板层素(LN),分析二者与AILTV致病力关系。结果显示:所分离的鸡传染性喉气管炎病毒中AILTV-HB-2不会导致LMH细胞贴壁力下降,其他毒株可导致LMH细胞贴壁力下降。AILTV-HB-1可导致LMH细胞LN蛋白降解,从而降低LMH细胞贴壁力,使其游离性增加,增强其增殖速度,推测温和型AILT病症气管中的干酪样堵塞物的产生可能与病毒降解LN有关。 Infectious laryngotracheitis virus(AILTV)can cause upper respiratory infections in chickens,which seriously endangers the development of the chicken industry.During the winter of 2017to the spring of 2018,we isolated and identified 8strains of AILTV from the chickens in the chicken farms in Hebei province,numbered AILTV-HB-1-AILTV-HB-8.These eight AILTV wild viruses and two vaccines and Wanggang strains were vaccinated separately into the chicken liver cancer cell(LMH)to detect the proliferation rate of LMH,while using ELISA to detect changes in laminin and integrin.The results showed that AILTV-HB-2in the isolated infectious laryngotracheitis virus did not decrease the adhesion of LMH cells,and other strains could cause the adhesion of LMH cells to decrease.AILTV-HB-1can cause LN protein degradation,thereby reducing the adhesion of LMH cells,increasing their freeness and increasing their proliferation rate.It is speculated that the formation of cheese-like plugs in trachea may be related to the degradation of LN.
