没食子酰芍药苷在脑缺血再灌注后神经保护作用和机制 被引量:2

The neuroprotective effects of galloyl paeoniflorin on cerebral ischemia-reperfusion and the underlying mechanism

摘要 Wistar大鼠通过线栓法建立脑缺血再灌注模型,给予不同剂量没食子酸芍药苷治疗。结果显示,没食子酰芍药苷治疗使大鼠脑梗死体积明显减小,神经功能评分降低,并诱导大鼠脑组织中氧化物歧化酶和谷胱甘肽过氧化物酶活性、谷胱甘肽含量以及Nrf2/HO-1表达明显升高,而半胱天冬酶3活性、丙二醛含量、肿瘤坏死因子α和白介素1β的表达以及Tunel染色阳性细胞数明显降低。结果表明:没食子酰芍药苷可激活Nrf2/HO-1通路,减轻脑缺血再灌注后氧化应激和炎症反应,从而起到神经保护作用。 Cerebral ischemia-reperfusion models of Wistar rats were established by line plug method,and treated with different doses of paeoniflorin.The results showed that galloyl paeoniflorin treatment significantly reduced the infarct volume of rats,decreased the neurological function score,and induced the activity of oxide dismutase,glutathione peroxidase and glutathione in rat brain tissue.The expression of Nrf2/HO-1was significantly increased,while the activity of caspase 3,malondialdehyde,tumor necrosis factor alpha and interleukin 1βand the number of Tunel staining positive cells were significantly decreased.The results showed that galloyl paeoniflorin can activate Nrf2/HO-1pathway,reduce oxidative stress and inflammatory response after cerebral ischemia-reperfusion,and thus play a neuroprotective role.
