摘要
采用初筛和复筛试验,从土壤中筛选获得一株产阿魏酸酯酶活力较高的菌株A3,经形态观察、生理生化特性结合18S/ITS功能区进行测序鉴定其为黄曲霉(Aspergillus flavus)。通过单因素轮换法和正交试验设计,对该菌株进行产酶条件考察及发酵条件优化,结果表明:在接种量5%、温度28℃、180 r/min条件下培养84 h,菌株A3产阿魏酸酯酶的酶活最高为843.52 mU/mL。
A strain A3 with high ferulase activity was screened from soil by primary screening and rescreening tests. The strain A3 was identified as Aspergillus flavus by morphological observation, physiological and biochemical characteristics and 18 S/ITS functional region sequencing. Then the fermentation conditions of the strain were optimized by single factor rotation method and orthogonal design. The results showed that the highest ferulic esterase activity of strain A3 was 843.52 mU/mL when inoculated at 5% inoculation, temperature 28 ℃, and 180 r/min for 84 h.
