摘要
研究使用多种生物信息学分析软件和多种在线生物信息分析工具,探究鸡成纤维细胞生长因子13(FGF13)基因及其编码蛋白的序列结构及遗传学特性,并分析其组织表达情况。结果显示:鸡FGF13基因编码区DNA序列长582 bp,共编码氨基酸193个;同源性分析发现,鸡FGF13基因具有较高的保守性,其核苷酸序列及其编码的氨基酸序列与鸟类中鸽和火鸡的同源性均达95%以上;氨基酸序列分析发现,该蛋白为不稳定的水溶性蛋白,分子质量为21.65 ku,理论等电点为9.33,无信号肽序列;亚细胞定位主要为细胞核内,属于非分泌蛋白;预测含有30个磷酸化位点,3个糖基化位点,具有FGF结构域;二级结构主要以无规则卷曲和β折叠为主,三级结构为卷曲状折叠结构;该基因在鸡早期生长过程中随周龄的增大在腿肌中的表达量呈下降模式。研究结果为FGF13基因功能的进一步分析,以及该基因在鸡生长发育及其他方面调控作用研究提供了更充分的分子生物学基础信息和理论依据。
This study was aimed to explore the sequence structure and genetic characteristics of chicken FGF13 gene and its encoded protein, and to analyze its expression during early growth and development, using multiple bioinformatics analysis software and online tools. The results showed that the length of the coding region of FGF13 gene was 582 bp,which encoded 193 amino acids. The homology analysis revealed that FGF13 gene of chicken was highly conserved,because its nucleotide sequence and its encoded amino acid sequence both shared more than 95% identity with pigeon and turkey in birds.The amino acid sequence analysis showed that FGF13 protein was unstable water-soluble protein with a molecular mass of 21.65 ku and its pI was 9.33, and had no signal peptide sequence.Subcellular localization was located at the nucleus, belonging to non-secreted protein.The FGF13 protein contained 30 phosphorylation sitesand 3 glycosylation sites, with FGF domain. The secondary structure of FGF13 was mainly composed of random coil and β-folding,and the tertiary structure was a coiled-folded mixed structure. The expression of FGF13 gene in muscle decreased with increasing age in the early growth of chicken.These results provided basic information of molecular biology and theoretical foundation for further study on the role and regulatory mechanism of this gene in chicken growth and other aspects.
