摘要
目的:探讨益胆汤防治胆囊胆固醇结石作用机制,分析SCF/c-Kit信号通路。方法:将60只豚鼠随机均等分为A、B、C三组。其中A组行常规饲料饲养,B组给予含高胆固醇的饲料进行饲养,C组行高胆固醇的饲料+益胆汤饲养。喂养6周后处死豚鼠以检测胆固醇结石生成情况。同时行Western blot法、RT-PCR法对小鼠胆组织中的c-Kit及SCF的含量进行检测。结果:A组成石率低于C组,且两组均低于B组(P<0.05);B组胆固醇浓度及胆汁酸浓度高于C组,且两组均高于A组(P<0.05),A组致石指数与C组较为一致,均显著低于B组(P<0.05);B组与A组豚鼠组织中,其c-Kit与SCF及GAPDH基因有效扩增,B组c-Kit与SCF的mRNA的表达明显低于A组(P<0.05)。而C组在c-Kit与SCF蛋白表达上明显高于B组,且均低于A组(P<0.05)。结论:益胆汤能够有效提高豚鼠体内SCF/c-Kit的表达,防治胆囊胆固醇结石效果较好。
Objective: To study the prevention and treatment of gallbladder cholesterol calculus mechanism beneficial gall soup, analysis of SCF/c-Kit signaling pathways. Method: The equality of 60 guinea pigs were randomly divided into A, B, C three groups. The number control line of conventional breeding, group B was given feed containing high levels of cholesterol in breeding, feed line high cholesterol group C + breeding benefit bile soup. Six weeks after feeding guinea pigs were executed to detect cholesterol calculus formation. Line and Western blot method, PT-PCR method for small dept c-Kit and SCF levels in the organization for testing. Result: The composition of A stone rate was lower than that of group C, and the two groups were lower than that of group B(P<0.05). Cholesterol and bile acid concentrations higher than group B, group C and two groups were higher than group A(P<0.05), group A and C of stone index were more consistent and significantly lower than that of group B(P<0.05). Group B and group A guinea pig in the organization, its c-Kit and SCF and GAPDH gene amplification effectively, the result shows, group B c-Kit with SCF mRNA expression were significantly lower than the control group(P<0.05). While C-Kit and SCF protein expression in group C were significantly higher in group B, and were lower than that of group A(P<0.05). Conclusion: The beneficial gall soup can effectively improve the expression of SCF/c-Kit in guinea pigs, the effect of prevention and treatment of gallbladder cholesterol calculus.
