Simultaneous determination of five components in Herba Artemisiae Anomalae by HPLC

摘要 目的:建立HPLC法同时测定刘寄奴中槲皮素、柚皮素、山柰酚、芒柄花素、异泽兰黄素5个成分的含量。方法:采用Syncronis C18色谱柱(250 mm×4.6 mm,5μm),流动相为甲醇-0.2%磷酸水(40:60),检测波长260 nm,流速1.0 mL·min-1,柱温25℃,进样量10μL。结果:将实验测得的图谱与标准物质进行对照,确定其5个成分分别为槲皮素、柚皮素、山柰酚、芒柄花素、异泽兰黄素;5个成分分别在测定范围内(0.062~0.992μg,0.031~0.496μg,0.034~0.544μg,0.022~0.352μg,0.027~0.432μg)线性关系良好;平均加样回收率为97.8%~100.6%,RSD均小于3.0%;4批样品中5个成分的含量分别为0.022%~0.041%、0.009%~0.024%、0.011%~0.023%、0.006%~0.014%、0.017%~0.033%。结论:该方法简便准确,重复性好,可用于刘寄奴5个成分的含量测定。 Objective:To establish an HPLC method for simultaneous determination of five components in Herba Artemisiae Anomalae.Methods:The HPLC assay was performed on a Syncronis C18(250 mm×4.6 mm,5 μm)column.The mobile phase was methanol-0.2% phosphoric acid(40:60).The flow rate was 1 mL·min-1,the detection wavelength was 260 nm,the column temperature was 25 ℃ and the injection volume was 10 μL.Results:Five constituents of quercetin,naringin,kaempferol,formononetin and eupatilin were identified by comparing with the spectra of standard substances.The linear relationship of the five components was good within the range(0.062-0.992 μg,0.031-0.496 μg,0.034-0.544 μg,0.022-0.352 μg and 0.027-0.432 μg).The average recoveries of sample addition were in the range of 97.8%-100.6%(RSD<3.0%).The contents of five components in four batches of samples were 0.022%-0.041%,0.009%-0.024%,0.011%-0.023%,0.006%-0.014% and 0.017%-0.033%.Conclusion:The method is simple,accurate and reproducible.It can be used for the determination of five components in Herba Artemisiae Anomalae.
