疏肝和胃丸对糖尿病胃轻瘫模型大鼠胃动力及胃肠激素的影响

Effect of Shugan Hewei pills on gastric motility and gastrointestinal hormones in a rat model of diabetic gastroparesis

目的:探讨疏肝和胃丸对糖尿病胃轻瘫模型大鼠胃动力及胃肠激素的影响。方法:制备糖尿病胃轻瘫模型大鼠,按体质量随机分为模型组、西药对照组和疏肝和胃丸高、中、低剂量组,疏肝和胃丸3个剂量组分别给予临床剂量1、2、4倍灌胃,西药对照组予临床剂量1倍的多潘立酮灌胃,另设正常对照组,与模型组均灌胃等体积蒸馏水。各组用药4周后检测胃排空率、肠推进率、胃蠕动功能、胃血流量、血清胃动素(MOT)、胃泌素(GAS)和胰高血糖素(GLG)含量。结果:与模型组相比,疏肝和胃丸高、中、低3个剂量组胃排空率、胃蠕动频率及幅度、胃血流量等指标均显著增高,血清MOT、GAS、GLG含量均显著降低,差异均有统计学意义(P <0. 05,P <0. 01)。结论:疏肝和胃丸能促进糖尿病模型大鼠胃动力,可能与增加胃血流量、改善胃肠组织缺血缺氧、调节胃肠激素功能有关。

Objective: To investigate the effect of Shugan Hewei pills on gastric motility and gastrointestinal hormones in a rat model of diabetic gastroparesis. Methods: A rat model of diabetic gastroparesis was established,and according to body weight,the rats were randomly divided into model group,Western medicine control group,and high-,middle-,and low-dose Shugan Hewei pills groups. The high-,middle-,and low-dose Shugan Hewei pills groups were given Shugan Hewei pills by gavage at a dose 1,2,and 4 times the clinical dose,and the Western medicine group was given domperidone by gavage at the same dose as the clinical dose. The normal control group and the model group were given an equal volume of distilled water by gavage. Gastric emptying rate,intestinal propulsion rate,gastric peristalsis function,gastric blood flow,and serum levels of motilin(MOT),gastrin(GAS),and glucagon(GLG) were measured after 4 weeks of administration. Results: Compared with the model group,the high-,middle-,and low-dose Shugan Hewei pills groups had significant increases in gastric emptying rate,frequency and amplitude of gastric peristalsis,and gastric blood flow and significant reductions in serum MOT,GAS,and GLG(P < 0. 05 or P < 0. 01).Conclusion: Shugan Hewei pills can improve gastric motility in the rat model of diabetes,possibly by increasing gastric blood flow,improving ischemia and hypoxia of gastrointestinal tissue,and regulating gastrointestinal hormone function.
