电针少阳经穴对偏头痛大鼠行为学、50%缩足阈值及血清ET、NO、CGRP含量的影响 被引量:23

Effects of electro-acupuncture at specific acupoint on Shaoyang meridian in different periods on behavior, 50% paw withdrawal threshold and serum ET, NO, CGRP contents in migraine rats

摘要 目的:观察不同时期电针少阳经穴对偏头痛大鼠的行为学、50%缩足阈值(50%PWT)及血清中内皮素(ET)、一氧化氮(NO)、降钙素基因相关肽(CGRP)含量的影响,为临床运用针灸治疗偏头痛提供合理依据。方法:将50只大鼠随机分为空白组、模型组、电针少阳经穴预防组、电针少阳经穴治疗组、电针少阳经穴预防+治疗组,每组10只。空白组第8天皮下注射等量0.9%氯化钠注射液,30min后捆绑30min;其余各组第8天皮下注射硝酸甘油造模,其中模型组、电针少阳经穴预防组造模30min后捆绑30min,电针少阳经穴治疗组、电针少阳经穴预防+治疗组造模30min后捆绑行电针治疗30min,每日1次,持续7d。观察造模前后及治疗后大鼠行为学和50%PWT,酶联免疫定法(ELISA)检测各组血清中ET、CGRP的含量,采用硝酸还原酶法检测各组血清中NO的含量。结果:与空白组比较,造模后模型组大鼠行为学评分明显升高(P<0.01),50%PWT明显降低(P<0.01);与模型组比较,电针少阳经穴各治疗组治疗后行为学评分明显降低(P<0.01),50%PWT明显升高(P<0.01),血清ET、NO、CGRP含量降低(P<0.01);与电针少阳经穴预防组、电针少阳经穴治疗组比较,电针少阳经穴预防+治疗组治疗后行为学评分明显降低(P<0.01),50%PWT明显升高(P<0.01),血清ET、NO、CGRP含量显著降低(P<0.01)。结论:电针少阳经穴可以通过下调ET、NO、CGRP等血管活性物质的表达,抑制神经源性炎性反应的产生和三叉神经血管系统的激活,从而减轻偏头痛的疼痛程度,而电针少阳经穴预防+治疗的方式对ET、NO、CGRP等血管活性物质的表达下调幅度最显著,缓解偏头痛的效果较单纯预防性治疗和单纯发作期治疗更优。 Objective: To observe the effects of electro-acupuncture at specific acupoints of Shaoyang Meridian in different periods on the behavior and the levels of 50% Paw withdrawal threshold(PWT), serum endothelin(ET), nitric oxide(NO) and calcitin gene-related peptide(CGRP) of migraine rats, so as to provide a reasonable basis for clinical application of acupuncture and moxibustion in the treatment of migraine. Methods: Fifty rats were randomly divided into five groups: control group, modelgroup, prevention group of electro-acupuncture at Shaoyang meridian, treatment group of electro-acupuncture at Shaoyang meridian, prevention plus treatment group of electro-acupuncture at Shaoyang meridian, with 10 rats in each group. The rats from prevention group of electro-acupuncture at Shaoyang meridian and prevention plus treatment group of electroacupuncture at Shaoyang meridian were bound and given the electro-acupuncture 30 min per day for 7 days, while other three groups were only bound at the same time. The control group was injected with the same volume of normal saline, and then bound for 30 minutes after 30 minutes, while the other groups were subcutaneously injected with nitroglycerin on the 8 th day. The model group and the electro-acupuncture prevention group of Shaoyang meridian were bound for 30 minutes after 30 minutes. The electro-acupuncture treatment group and the electro-acupuncture prevention plus treatment group were bound for 30 minutes after modeling. Behavior and 50% PWT of rats were observed before and after the establishment of models. The levels of ET and CGRP in serum were detected by enzyme-linked immunosorbent assay(ELISA). Furthermore, the level of NO in serum was detected by nitrate reductase method. Results: Compared with the control group, the behavioral score of rats in the model group increased significantly(P<0.01), and the 50% PWT decreased significantly(P<0.01). Compared with the model group, the behavioral score of rats in the electro-acupuncture groups at Shaoyang meridian decreased significantly(P<0.01), the 50% PWT increased significantly(P<0.01), and the contents of serum ET, NO and CGRP decreased significantly(P<0.01). Compared with Shaoyang meridian prevention group and Shaoyang meridian treatment group, the behavioral score of Shaoyang meridian prevention plus treatment group decreased significantly(P<0.01), 50% PWT increased significantly(P<0.01), and serum ET, NO, CGRP decreased significantly(P<0.01). Conclusion: Electro-acupuncture at specific acupoint of Shaoyang meridians can reduce the expression of vascular active substances, such as ET, NO and CGRP,inhibiting the generation of neurogenic inflammation and the activation of trigeminal vascular system, thereby reducing the level of migraine pain.The prevention plus treatment group of electro-acupuncture at Shaoyang meridian was the most significant decline in the expression of vascular active substances, which relieved migraine better than in preventive treatment or simple acute phase therapy.
