血红栓菌液体发酵产漆酶培养条件的研究 被引量:3

摘要 采用液体培养,考察碳源、氮源、Cu^2+及其他培养条件对血红栓菌产漆酶的影响。采用2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐法测定血红栓菌菌株所产漆酶的酶活力。结果表明,以葡萄糖15 g·L^-1和蔗糖15 g·L^-1为碳源,蛋白胨2 g·L^-1和硝酸铵2 g·L^-1为氮源,CuSO42.0 mmol·L^-1,初始pH 5.0,300 mL三角瓶装液量为100 mL,接种量8%,温度28℃,在此条件下,发酵液漆酶酶活为165.19 U·mL^-1,比优化前的53.80 U·mL^-1提高了3倍。 The effect of carbon resource,nitrogen source,Cu^2+ and other culture conditions on laccase by liquid fermentation cultivation of the Trametes sanguinea were studied.The laccase production of T.sanguinea was detected by 2,2’-azinobis-(3-ethylbenzthiazoline-6-sulphonate)method.The results showed that,the optimum conditions were glucose 15 g·L^-1,sucrose 15 g·L^-1,peptone 2 g·L^-1,ammonium nitrate 2 g·L^-1,CuSO42.0 mmol·L^-1,and the initial pH was 5.0,liquid medium volume was100 mL per 300 mL,the inoculation quantity was 8%and the temperature was 28℃.Under these conditions,the maximal laccase activity was 165.19 U·mL^-1,which was 3 times higher than that before optimization(53.80 U·mL^-1).
