2 633例老年汉族患者SLCO1B1及ApoE基因多态性分析

Gene polymorphisms of SLCO1B1 and ApoE in 2 633 Chinese Han elderly patients

目的探讨老年汉族患者溶质载体有机阴离子转运蛋白家族1B1(SLCO1B1)和载脂蛋白E(ApoE)基因多态性的分布情况,为老年人合理用药提供依据。方法采用聚合酶链反应-荧光探针方法对2016年4月-2018年4月解放军总医院第四医学中心2633例心血管疾病患者的SLCO1B1和ApoE基因多态性进行检测,纳入对象年龄≥60岁,其中男性1427例,女性1206例,分析基因型分布的性别差异。结果SLCO1B1基因的*1a/*1a+*1a/*1b+*1b/*1b组合最多见,占78.28%(2061/2633);*5/*5+*5/*15+*15/*15组合33例,占1.25%。各基因型组合的性别分布差异无统计学意义(P>0.05)。ApoE基因的E2/E4+E3/E3组合最多见,占69.69%(1835/2633),E3/E4+E4/E4组合有430例,占16.33%,各基因型组合的性别分布差异无统计学意义(P>0.05)。结论本研究展示了大样本老年汉族人群中SLCO1B1及ApoE基因多态性的分布,结果对调整的用药方案具有重要意义。

Objective To investigate the distribution of genetic polymorphisms of SLCO1B1 and ApoE in Chinese Han elderly patients,so as to provide evidences for rational use of drugs in elderly patients.Methods The genetic polymorphisms of SLCO1B1 and ApoE in 2633 patients with cardiovascular disease admitted to our center from April 2016 to April 2018 were detected by the PCR fluorescence probe technique.The age of the included patients were all greater than 60 years,with 1427 males and 1206 females.Results In SLCO1B1 genotype,the frequency of*1a/*1a+*1a/*1b+*1b/*1b was 78.28%(2061/2633),followed by*1a/*5+*1a/*15+*1b/*15(539,20.47%)and*5/*5+*5/*15+*15/*15(33,1.25%);The distribution of each genotype combination had no statistically significant difference between male and female(P>0.05).The frequency of ApoE E2/E4+E3/E3 was 69.69%(1835/2633),followed by the combination of ApoE E3/E4+E4/E4(430,16.33%),and E2/E2+E2/E3(368,13.98%);The distribution of each genotype combination had no statistically significant difference between male and female(P>0.05).Conclusion Our study demonstrate gene polymorphisms of SLCO1B1 and ApoE in elderly people,which is beneficial for prescribing appropriately in the elderly populations.
