大柴胡汤对2型糖尿病模型大鼠血糖的影响 被引量:15

摘要 目的:观察大柴胡汤对2型糖尿病(T2DM)模型大鼠空腹血糖(FBG)的影响,探讨该方防治T2DM的作用机理。方法:60只雄性SD大鼠适应性喂养1周后,随机抽出12只为对照组,其余以高脂饮食喂养8周后,一次性腹腔注射链脲佐菌素(STZ)建立T2DM模型。随机选择造模成功的大鼠36只,分成模型组、大柴胡汤(8.1g/kg)组、二甲双胍(50mg/kg)组,其中,大柴胡汤组、二甲双胍组分别给予药物灌胃,1次/d,对照组和模型组给予相同生理盐水灌胃;8周后,检测大鼠FBG、血清总胆固醇(TC)、甘油三酯(TG)、游离脂肪酸(FFA)和空腹胰岛素(FINS),计算胰岛素抵抗指数(HOMA-IR)。实验结束后,迅速处死大鼠,检测大网膜脂肪组织磷脂酰肌醇-3激酶(PI3K)、磷酸化蛋白激酶B(P-AKT)、葡萄糖转运蛋白4(GLUT4)的蛋白表达。结果:与模型组比较,大柴胡汤组FBG、TC、TG、FFA、FINS、HOMA-IR明显降低(P<0.01),大网膜脂肪组织PI3K、P-AKT、GLUT4蛋白表达均明显上升(P<0.01)。结论:大柴胡汤能降低T2DM模型大鼠血清FBG、TC、TG、FFA;改善IR,其机制可能与激活PI3K/AKT信号通路,增加葡萄糖的运转有关。 Objective:To observe the effect of Dachaihu decoction on fasting blood glucose(FBG)in type 2 diabetes mellitus(T2DM)rats,and to explore the mechanism of prevention and treatment of T2DM.Methods:One week after adaptive feeding of 60 male Sprague-Dawley rats,12 rats were randomly selected as control group.After 8 weeks of feeding on high-fat diet,T2DM model was established by intraperitoneal injection of streptozotocin(STZ).Thirtysix rats with successful modeling were randomly divided into model group,Dachaihu decoction(8.1g/kg)group and metformin(50mg/kg)group.Among them,Dachaihu decoction group and Metformin group were given drugs,1 Times/d,the control group and the model group were given the same saline;after 8 weeks,the rats were tested for FBG,serum total cholesterol(TC),triglyceride(TG),free fatty acid(FFA)and fasting insulin(FINS).,Calculate the insulin resistance index(HOMA-IR).At the end of the experiment,the rats were quickly sacrificed and the protein expression of retinoic acid inositol-3 kinase(PI3K),phosphorylated protein kinase B(P-AKT),and glucose transporter 4(GLUT4)was detected.Results:Compared with the model group,the FBG,TC,TG,FFA,FINS,and HOMA-IR of the Dachaihu decoction group were significantly decreased(P<0.01),and the expression of PI3K,P-AKT,and GLUT4 proteins in the omental adipose tissue were significantly increased(P<0.01).Conclusion:Dachaihu decoction can reduce serum FBG,TC,TG and FFA in T2DM model rats.The mechanism of IR may be related to the activation of PI3K/AKT signaling pathway and the increase of glucose.
