都匀毛尖茶树叶片响应茶小绿叶蝉侵染的转录组分析 被引量:3

Transcriptome Analysis of the Infection of the Leaves of Duyun Maojian Tea in Response to Green Leafhopper

摘要 茶小绿叶蝉是为害茶树叶片主要的病害之一,可造成茶叶显著减产。为了研究都匀毛尖茶对茶小绿叶蝉侵染响应的分子机制,通过Illumina Hiseq平台测序,对茶小绿叶蝉侵染茶树叶片转录组基因的表达情况进行分析。结果表明,从9个样品筛选出的SNP位点数目从490103~632564,并对214个基因结构进行了优化,发现了13588个新基因,其中10183个新基因被注释。茶小绿叶蝉侵染0 h和12 h两组出现了555个差异基因,其中上调表达369个,下调表达186个。0 h和24 h两组出现了1575个差异转录基因,677个上调,898个下调。12 h和24 h两组出现了22个差异表达基因,4个上调,18个下调。将差异表达基因进行COG、GO、KEGG、KOG、NR、Pfam、Swiss-Prot和egg NOG注释,并通过GO和KEGG功能分类和富集分析发现,差异基因主要参与植物激素信号转导、应激反应,解毒反应,免疫系统过程。研究结果为揭示茶叶响应茶小绿叶蝉侵害的防御机制、关键基因提供理论依据,对于选育抗性良种并最终实现茶叶增产意义重大。 Tea lesser leafhopper is one of the major pests in tea,which can significantly reduce the yield of tea.In order to study the molecular mechanism of tea infection response to tea lesser leafhopper,the expression of transcriptome genes in tea infected by green leafhopper was analyzed by Illumina Hiseq platform sequencing.The results showed that SNP number was from 490103 to 632564 among the nine samples,214 gene structures were optimized and 13588 new genes were found.There were 555 DEGs appeared between 0 h and 12 h after the infection of tea lesser leafhopper,369 genes were up-regulated and 186 were down-regulated.1575 DEGs were found between 0 h and 24 h,677 genes were up-regulated and 898 were down-regulated.22 DEGs were got between 12 h and 24 h,4 genes up-regulated and 18 genes were down-regulated.DEGs were annotated by COG,GO,KEGG,KOG,NR,Pfam,Swiss-Prot and egg NOG.Also,DEG mainly involving in plant hormone signal transduction,response to stimulus,detoxification,immune system process.The results provide a theoretical basis for revealing the defense mechanism and key genes of tea in response to tea green leaf mites,and it is of great significance for breeding resistant varieties and ultimately achieving tea yield increase.
