海南桑树种质资源SSR标记的遗传多样性分析 被引量:1

摘要 为了明确海南桑树种质资源间的亲缘关系,本研究利用SSR标记在分子水平上对30份海南桑树种质资源进行遗传多样性分析。试验从30对SSR引物中筛选出10对有效引物,共扩增出170条谱带,其中148条为多态性条带,多态位点百分率(PPB)为87.18%,个体间的遗传相似系数(GS)变化范围为0.442~0.89,平均GS值为0.6045,推论得知桑树引进资源与海南本地种质资源间有着较远的亲缘关系。通过POPGENE1.32软件计算海南桑树遗传多样性。结果表明:在物种水平上,有效等位基因数Ne=1.5150,Nei’s基因多样性H=0.3000,Shannon信息指数I=0.4490。基于遗传相似系数采用UPGMA法构建了30份海南桑树种质资源的遗传关系系统树,在遗传相似系数为0.578时可将全部种质分为2个类群,且该聚类方式与地理位置密切相关。本研究基于SSR分子标记分析海南桑树种质资源遗传多样性并进行亲缘关系的聚类,从DNA分子水平上揭示海南桑树的遗传关系,为海南桑树种质创新利用提供理论依据。 In order to clarify the genetic relationship between mulberry germplasm resources in Hainan,the genetic diversity of 30 mulberry germplasm resources from Hainan were analyzed at the molecular level by simple sequence repeat(SSR)markers.Ten pairs of effective primers were selected from 30 pairs of SSR primers,and a total of 170 bands were amplified,and 148 bands were polymorphic.The percentage of polymorphic loci(PPB)was 87.18%.The genetic similarity coefficient(GS)between individuals varies from 0.442 to 0.89,and the average GS value is 0.6045,it is inferred that there is a long-distance relationship between the introduction of mulberry trees and the local germplasm resources in Hainan.The genetic diversity of mulberry in Hainan was calculated by POPGENE 1.32 software.The results showed that at the species level,the number of effective alleles(Ne)per locus was 1.5150,the expected heterozygosity(He)was 0.3000,and the Shannon diversity index(I)was 0.4490.Based on the genetic similarity coefficient,30 genetic relationships system trees of Hainan mulberry germplasm resources were constructed by UPGMA method.When the genetic similarity coefficient is 0.578,all germplasm can be divided into two groups,and the clustering results are correlated with the geographical location.Based on SSR molecular markers,the genetic diversity of mulberry germplasm resources in Hainan was analyzed and clustered.The genetic relationship of mulberry trees in Hainan was revealed from the molecular level of DNA,which provided a theoretical basis for the innovative utilization of mulberry germplasm in Hainan.
