人巨细胞病毒感染对单核细胞功能的影响 被引量:2

Effect of Infection by the Human Cytomegalovirus on Monocyte Function

摘要 人巨细胞病毒(Human cytomegalovirus,HCMV)是一种β疱疹病毒。单核细胞是HCMV的主要体内靶标。当前大多数的研究集中于单个时间点来分析HCMV感染对单核细胞功能的影响,并未整体且动态地阐述HCMV感染对单核细胞功能的影响。本研究从GEO(Gene Expression Omnibus)数据库筛选HCMV感染单核细胞4h、24h、48h后的基因芯片,通过生物信息学方法对HCMV感染单核细胞4h、24h、48h后的功能影响进行详细的阐述及比较分析,并用定量反转录聚合酶连锁反应(quantificational reverse transcription polymerase chain reaction,qRTPCR)验证HCMV感染人单核细胞白血病细胞(THP-1细胞)4h、24h和48h后白细胞介素(Interleukin,IL)-1β、IL-8、肿瘤坏死因子(Tumor necrosis factor,TNF)-a、IL-10的表达。结果表明HCMV感染单核细胞4h后参与信号转导、炎症反应、免疫反应、防御病毒反应和趋化反应等17个通路,且可能与病理学过程/疾病、病毒疾病、RNA病毒感染和免疫系统疾病等相关。HCMV感染单核细胞24h后主要参与炎症反应、细胞趋化等信号通路,预测主要与系统性红斑狼疮(Systemic lupus erythematosus,SLE)、结缔组织疾病和自身免疫性疾病等10种疾病相关。HCMV感染单核细胞48h后主要参与炎症反应、细胞趋化等信号通路,预测主要与结缔组织疾病、病理学过程和SLE等10个疾病相关。qRT-PCR结果显示HCMV感染THP-1细胞4h、24h和48h后IL-1β、IL-8、TNF-α、IL-10的表达增加。本研究表明HCMV感染主要引起炎症反应及趋化反应,并可能与自身免疫性疾病或SLE高度相关。 The human cytomegalovirus(HCMV)is a beta herpesvirus. Monocytes are the major targets of the HCMV in vivo. Most studies have focused on the effects of the HCMV infection on monocyte function at a single time point. The effect of the HCMV infection on monocyte function has not been described in a comprehensive or dynamic manner. We screened the gene chips of HCMV-infected monocytes at 4 hrs,24 hrs and 48 hrs post infection from the GEO database. We investigated the functional effects of HCMV-infected monocytes at 4 hrs,24 hrs and 48 hrs post infection by bioinformatics analyses. The expression of interleukin(IL)-1β,IL-8,tumor necrosis factor(TNF)and IL-10 was detected by quantitative reverse transcription-polymerase chain reaction(qRT-PCR) in HCMV-infected THP-1 cells at 4 hrs,24 hrs and 48 hrs post infection. HCMV-infected monocytes were involved in 17 pathways(e.g.,signal transduction,inflammatory response,immune response,defense against viral response and chemotactic response)and may be associated with pathologic processes,viral diseases,RNA viral infections and immune-system diseases. At 24 hrs post infection,HCMV-infected monocytes were mainly involved in the inflammatory response,cell chemotaxis and other signaling pathways,and were mainly related to 10 diseases,including systemic lupus erythematosus(SLE),connective-tissue diseases and autoimmune diseases. At 48 hrs post infection,HCMV-infected monocytes were mainly involved in signaling pathways such as inflammation and cell chemotaxis. The prediction was mainly related to 10 diseases,such as connective-tissue disease,pathologic processes,and SLE. The results of qRT-PCR showed that the expression of IL-1β,IL-8,TNF-α and IL-10 increased in HCMV-infected THP-1 cells at 4 hrs,24 hrs and 48 hrs post infection. The present study suggested that HCMV infection could cause inflammatory and chemotactic reactions and may be highly correlated with autoimmune diseases or SLE.
