摘要
目的通过建立大鼠急性痛风性关节炎(AGA)模型,探讨白细胞介素17(IL-17)和核因子κB(NF-κB)在AGA中的表达及其意义。方法 45只SD大鼠按随机数字表法分为对照组(9只)和模型组(分为6、12、24 h及72 h组,每组9只)。将尿酸钠注射于大鼠右后踝关节腔内建立大鼠AGA模型,对照组注射等体积生理盐水,模型组各亚组分别在造模后6、12、24、72 h后处死,采用酶联免疫吸附法检测血清及滑膜中白细胞介素1β(IL-1β)、肿瘤坏死因子-α(TNF-α)和IL-17的表达,采用免疫印迹法检测滑膜中NF-κB的表达。结果与对照组比较,造模后6 h,模型组大鼠IL-1β、TNF-α和IL-17表达明显升高(P <0.01),12 h达到顶峰,24 h开始下降;模型组滑膜中p-NF-κB p65蛋白的相对表达水平均显著高于对照组(P <0.01),且随时间进展而逐渐下调。结论 AGA后,IL-17及p-NF-κB p65的表达水平显著增高,炎性反应加重,IL-17和NF-κB的差异表达与AGA的发生发展密切相关。
Objective To investigate the expression and significance of interleukin-17(IL-17) and nuclear factor kappa-B(NF-κB) in the acute gouty arthritis(AGA) rat model. Methods A total of 45 SD rats were divided into control group(9 rats) and model group(including four subgroups: 6, 12, 24 h and 72 h, 9 rats in each group) by random number table. A rat model of AGA was induced with the injection of monosodium urate(MSU) into the right posterior ankle joint cavity. The rats of the control group were injected with an equal dose of saline. The model group was sacrificed at6, 12, 24 h and 72 h, respectively. The expression of interleukin-1β(IL-1β), tumor necrosis factor-α(TNF-α) and interleukin-17(IL-17) in serum and synovium was detected by the enzyme-linked immunosorbent assay. The expression of NF-κB in synovial membrane was detected by Western blot. Results Compared with the control group, the expression of IL-1β, TNF-α and IL-17 in the model group was significantly increased at 6 h after AGA(P < 0.01), peaked at 12 h, and began to decrease at 24 h. The relative expression level of p-NF-κB p65 in the synovial membrane of the model group was significantly higher than that of the control group, and gradually decreased with time. Conclusion After AGA, the expression levels of IL-17 and p-NF-κB p65 are significantly increased, and the inflammatory response is aggravated. The differential expression of IL-17 and NF-κB is closely related to the occurrence and development of AGA.
