心脉隆注射液对阿霉素小鼠心脏损伤的保护作用及其机制 被引量:2

摘要 目的探讨心脉隆注射液对阿霉素小鼠心脏损伤的保护作用及其机制。方法将C57BL/6小鼠随机分为对照组10只,腹腔注射等体积生理盐水7d;阿霉素组10只,腹腔注射阿霉素10 mg/kg 2d+腹腔注射等体积生理盐水5d;心脉隆+阿霉素组10只,腹腔注射阿霉素10 mg/kg 2d+腹腔注射心脉隆注射液250 mg/kg 5d。7d后收集血清和组织标本,HE染色观察心脏组织病理改变、全自动生化分析检测心肌酶学指标变化、TUNEL和免疫组化分别观察心肌组织凋亡和自噬情况。结果阿霉素处理后小鼠出现心脏损伤表现,HE染色显示心肌纤维细胞和肌节结构紊乱、局部炎症细胞浸润、血清心肌酶学指标LDH和CK?MB升高(P<0.05)、心肌组织凋亡和自噬程度均增加(P<0.05);与阿霉素组相比,心脉隆处理能减轻心脏组织病理损伤、降低血清心肌酶学指标LDH和CK?MB(P<0.05)、减弱心肌组织细胞凋亡和自噬程度(P<0.05)。结论心脉隆注射液能够通过减轻心肌组织细胞凋亡和自噬水平,发挥对阿霉素小鼠心脏损伤的保护作用。 Objective To investigate the effect of Xinmailong injection against doxorubicin?induced cardiac injury and its mechanism in mice.Methods A total of 30 C57BL/6 mice were randomly divided into three groups:CTRL group(mice undergoing sham injection),DOX group(mice undergoing 10 mg/kg doxorubicin injection for 2 day)and XML+DOX group(mice undergoing 10 mg/kg doxorubicin injection for 2 day and 250 mg/kg Xinmailong injection for 5 day).After 7 days,Serum and heart tissue samples were collected.HE stain was used to analyze the histopathological changes in hearts.The myocardial enzymes were determined by Automatic biochemistry instrument.TUNEL and immunohistochemistry were respectively used to detect apoptosis and autophagy.Results Doxorubicin?treated mice occurred cardiac injury,which showed myocardial fibroblasts and sarcomeric structural disorders and inflammatory cell infiltration.Myocardial enzymes including LDH and CK?MB were increased(P<0.05),the levels of apoptosis and autophagy in cardiac tissue were both increased(P<0.05).Xinmailong injection could mitigate doxorubicin?induced histopathological abnormalities and elevated myocardial enzymes(P<0.05),meanwhile attenuate the levels of apoptosis and autophagy(P<0.05).Conclusion The study indicates Xinmailong injection could attenuate doxorubicin?induced cardiac injury and dysfunction,and reduce apoptosis and autophagy.
