摘要
目的探讨超声靶向微泡爆破技术对糖尿病肾病大鼠造模时间及肾功能的影响。方法取32只SD雄性大鼠,随机分为对照组(正常大鼠)、A组(给予链脲佐菌素建立糖尿病肾病大鼠模型)、B组(给予链脲佐菌素+超声微泡技术建立糖尿病肾病大鼠模型)、C组(给予链脲佐菌素+超声靶向微泡爆破技术建立糖尿病肾病大鼠模型),每组各8只。比较各组造模时间、尿蛋白排泄率(UAER)、空腹血糖(FBG)、血肌酐(SCr)、血尿素氮(BUN)、丙氨酸氨基转移酶(ALT)水平的差异。取各组大鼠肾脏组织,行苏木精-伊红染色,在光学显微镜下观察大鼠肾小球病理形态学改变。结果A、B、C组均成功建立糖尿病肾病大鼠模型,造模期间无大鼠死亡的情况发生。相比A、B组,C组造模时间明显减少(P<0.01);A、B组造模时间比较,并无显著差异(P>0.05)。相比对照组,A、B、C组UAER、FBG、SCr及BUN水平均明显升高(P<0.01);而各组ALT水平的比较,并无显著差异(P>0.05)。经病理染色结果可知,对照组大鼠肾脏组织未见异常病理改变,而A、B、C组大鼠均出现一系列不同程度的糖尿病肾病的病理改变。结论采用超声靶向微泡爆破技术制备糖尿病肾病大鼠模型可明显减少造模时间,具有良好的可行性和安全性。
Objective To analyze the effect of ultrasound targeted microbubble destruction on modeling time and renal function in diabetic nephropathy rats.Methods Thirty-two SD male rats were randomly divided into control group(normal rats),group A(diabetic nephropathy rat models established by streptozotocin),group B(diabetic nephropathy rat models established by streptozotocin+ultrasound microbubble technology),and group C(diabetic nephropathy rat models established by streptozotocin+ultrasound targeted microbubble destruction technology),8 rats in each group.The time of modeling,urinary albumin excretion rate(UAER),fasting blood glucose(FBG),serum creatinine(SCr),blood urea nitrogen(BUN),alanine aminotransferase(ALT)levels were compared among the groups.The pathological changes of rat glomeruli were observed under light microscope by haematoxylin-eosin staining.Results Diabetic nephropathy rat models were successfully established in groups A,B and C.No death occurred during the establishment of the models.Compared with group A and B,the modeling time of group C was significantly reduced(P<0.01),and there was no significant difference between group A and group B(P>0.05).Compared with the control group,UAER,FBG,SCr and BUN levels in group A,B and C were significantly higher(P<0.01),while there was no significant difference in the level of ALT among each group(P>0.05).The results of pathological staining showed that no abnormal pathological changes were found in the kidney tissues of rats in the control group,but a series of pathological changes of diabetic nephropathy appeared in groups A,B and C.Conclusion The establishment of diabetic nephropathy rat model by ultrasound targeted microbubble destruction can significantly reduce the time of modeling,and has good feasibility and safety.
