蒲公英水提液抗病毒有效部位筛选及体外抗病毒作用观察 被引量:11

摘要 目的筛选蒲公英水提液抗病毒有效部位,并探讨其体外抗病毒机制。方法通过蒸馏水回流提取法制备蒲公英水提液(样品A),采用水提醇沉法得到上清液部位(样品B)和沉淀部位(样品C),进一步将样品C通过大孔吸附树脂吸附、洗脱液洗脱,得到多个洗脱部位的纯化样品(水X部位、25%乙醇X部位、50%乙醇X部位、75%乙醇X部位,X为洗脱次序)。采用呼吸道合胞病毒(RSV)-MA104、单纯疱疹病毒(HSV)-MA104、肠道EV-71型病毒(EV-71)-MA104细胞感染模型及样品A筛选出EV-71病毒进行蒲公英样品抗病毒有效部位的筛选。样品B、C分别进行体外抗EV-71病毒实验,筛选出最有效部位后,并将其作为研究样本进一步探究蒲公英体外抗EV-71病毒的作用。结果样品C经50%乙醇醇沉后,沉淀部位溶解,经D101型大孔吸附树脂吸附,水1洗脱部位的TI值为73.52。抗病毒作用实验中的预防作用组TI值为24.25,阻断作用组的TI值为5.28。两组比较,P<0.05。结论蒲公英水提液抗EV-71病毒最有效部位为样品C经50%乙醇醇沉后的水1洗脱部位。蒲公英体外抗EV-71病毒的作用主要为预防作用。 Objective The effective antiviral components of water extract of dandelion were screened and the antiviral mechanism was preliminarily explored.Methods Dandelion water solution(sample A)was prepared by distilled water reflux extraction method,and we used the water extract-alcohol precipitation to get the supernatant fluid(sample B)and precipitation part(sample C),further,the sample C was adsorbed and eluted by macroporous adsorption resin adsorption and eluent to get the purified samples from different parts(water X parts,25%ethanol X parts,50%ethanol X parts,75%ethanol X parts,X for the elution order).Cell infection models of respiratory syncytial virus(RSV)-MA104,herpes simplex virus(HSV)-MA104,intestinal EV-71 virus(EV-71)-MA104 and sample A were used to screen EV-71 virus for the screening of effective antiviral sites in dandelion samples.Samples B and C were respectively tested for anti-EV-71 virus in vitro.After selecting the most effective site,they were used as study samples to further explore the anti-EV-71 virus effect of dandelion in vitro.Results After sample C was sunk by 50%ethanol,the precipitation part was dissolved and absorbed by D101 macroporous adsorption resin.TI value of water 1 elution part was 73.52.In the antiviral effect experiment,the TI value of the prevention group was 24.25,and the TI value of the blocking group was 5.28.TI value of the prevention group was greater than that of blocking group(P<0.05).Conclusions The most effective part of dandelion water extract against EV-71 virus is the water-1 elution part after sample C 50%ethanol precipitation.The main function of dandelion against EV-71 virus in vitro is prevention.
