变应性鼻炎miRNA图谱构建及分子标记物筛查研究 被引量:3

摘要 目的通过构建变应性鼻炎(AR)微小RNA(miRNA)图谱,筛选出差异表达miRNAs,进行分子标记物筛查。方法选择新疆医科大学第一附属医院耳鼻喉科住院的诊断为鼻中隔偏曲合并有变应性鼻炎(AR)患者3例为AR组,选取同期住院的鼻中隔偏曲不合并AR患者3例为非AR组。2组患者术前行鼻内镜检查与特异性过敏原检测,均在全麻下行鼻内镜下鼻中隔偏曲矫正手术,术中取部分中鼻甲黏膜。从鼻腔黏膜标本中成功提取RNA,采用Illumin第二代测序技术对miRNA文库进行高通量测序。结果检测共有1405个miRNA在AR和非AR鼻黏膜组织中共同表达,其中AR组2564个,非AR组2807个。样本间差异表达的miRNA有hsa-miR-144-3p、hsa-miR-223-3p、hsa-miR-6087、hsa-miR-142-5p、hsa-miR-142-5p、hsa-miR-142-5p等25个,差异表达的miRNA参与了胞吞作用、组氨酸代谢、收集导管酸分泌、突触小泡循环、哮喘5条通路。以差异表达的hsa-miR-3687为例,hsa-miR-3687的靶基因有35个。结论该研究筛选出的25个样本间差异表达的miRNA和差异表达的miRNA参与的5条通路,为AR的研究提供进一步实验的基础。 ObjectiveTo screen differential microRNA(miRNA)by constructing miRNA maps of allergic rhinitis and to identify molecular markers related to allergic rhinitis.MethodsThe patients,totaling 25,hospitalized in the department of Otolaryngology of the First Affiliated Hospital of Xinjiang Medical University,was selected as the case of nasal septum deviation and allergic rhinitis(AR).Three patients were selected as the AR group.Three patients of nasal septum deviation without allergic rhinitis(AR)were selected as the non-AR group.Two groups of patients underwent endoscopic examination and specific allergen test.All patients underwent endoscopic septal surgery under general anesthesia.Part of the middle turbinate mucosa was taken during operation.RNA was successfully extracted from nasal mucosa specimens,and high-throughput sequencing of miRNA libraries was performed using Illumin′s sequencing technology.ResultsA total of 1405 differential miRNAs were detected in AR and non-AR nasal mucosa tissues,including 2564 in the AR group and 2807 in the non-AR group.There were 25 differentially expressed miRNAs such as:hsa-miR-144-3p,hsa-miR-223-3p,hsa-miR-6087,hsa-miR-142-5p,hsa-miR-142-5p,hsa-miR-142-5p,etc.,differentially expressed miRNAs are involved in five pathways:endocytosis,histidine metabolism,collecting ductal acid secretion,and small synapses.Synaptic vesicle cycle,Asthma.Taking the differentially expressed hsa-miR-3687 as an example,there are 35 target genes of hsa-miR-3687.ConclusionThe study screened differentially expressed miRNAs between 25 samples,and differentially expressed miRNAs involved in five pathways,thereby providing a basis for further experiments in AR studies.
