天麻素对CUMS大鼠抑郁样行为及学习记忆功能的影响 被引量:3

摘要 目的:观察天麻素对慢性不可预见性温和应激(CUMS)大鼠抑郁样行为及学习记忆功能的影响。方法:雄性SD大鼠随机分为正常组、模型组、生理盐水组、舍曲林组(10 mg/kg,qd)、天麻素低、高剂量组(100 mg/kg、200 mg/kg,qd),除正常组外均接受CUMS造模,给治疗组2周的药物腹腔注射,生理盐水组注射等容量生理盐水。药物干预后,用糖水偏好实验、旷场实验、高架十字迷宫实验、强迫游泳实验评估大鼠行为,用Morris水迷宫实验评估大鼠学习记忆功能。结果:①与正常组相比,模型组大鼠的体质量、糖水偏好度、水平跨格次数、修饰次数、开放臂进入次数比例均降低,且强迫游泳静止不动时间延长(P<0.05或P<0.01);生理盐水组和模型组上述指标均相似(P>0.05);与生理盐水组相比,舍曲林组、天麻素低、高剂量组上述指标显示出部分恢复(P<0.05或P<0.01)。②Morris水迷宫实验正常组、舍曲林组、天麻素低、高剂量组大鼠靶象限活动时间百分比高于模型组和生理盐水组(P<0.05或P<0.01),且第3天舍曲林组、天麻素低、高剂量组逃避潜伏期均短于模型组(P<0.05或P<0.01)。结论:天麻素可缓解CUMS致抑郁症大鼠抑郁样行为,并对其学习记忆功能可能具有一定的改善作用。 Objective:To observe the effects of gastrodin on depression-like behaviors and learning and memory functions of rats with chronic unpredictable mild stress(CUMS).Methods:Male Sprague-Dawley(SD)rats were randomized into normal group,model group,saline group,sertraline group(10 mg/kg,ip,qd),and low and high dose gastrodin groups(100 mg/kg;200 mg/kg,ip,qd).All groups received CUMS modeling except the normal group.Rats in the treatment groups were intraperitoneally administered with the drugs for 2 weeks,and those in the saline group were treated with equivalent volume of saline.After drug intervention,the behaviors of rats were evaluated by sucrose preference,open field,elevated plus-maze,and forced swimming tests,and the learning and memory functions of rats were evaluated by Morris water maze test.Results:①Rats in the model group had decreased body weight,sucrose preference percentage,numbers of squares crossed and self-grooming as well as open arm entry frequencies,and lengthened immobility time in forced swimming test as compared to rats in the normal group(P<0.05 or P<0.01).The above indexes were similar in rats in saline group to those in model group(P>0.05).Compared with the saline group,the above indexes showed partial recovery in the sertraline group,the low and high dose gastrodin groups(P<0.05 or P<0.01);②Morris water maze test showed that the time percentage for target quadrant activity in normal,sertraline,low and high dose gastrodin groups was higher than that of model group and saline group(P<0.05 or P<0.01).And on the third day,the escape latency of rats in sertraline group,low and high dose gastrodin groups was shorter than that of model group(P<0.05 or P<0.01).Conclusion:Gastrodin can relieve depression-like behaviors of depressive rats induced by CUMS,and improve the learning and memory functions of such model rats as well.
