摘要
目的探讨长链非编码RNA(LncRNA)SNHG1对非小细胞肺癌(NSCLC)细胞迁移和侵袭的影响。方法利用GEO数据库数据集资料分析SNHG1在肺癌组织与正常组织的表达,同时分析SNHG1表达量与肺癌患者总生存期的关系;过表达及干扰NSCLC细胞中SNHG1的表达,qRT-PCR检测SNHG1的表达效率,细胞划痕实验、transwell迁移及侵袭实验分别检测SNHG1对NSCLC细胞迁移和侵袭的影响。通过蛋白质印迹法检测上皮细胞标志蛋白E-钙黏蛋白(E-cadherin)、间质细胞标志蛋白N-钙黏蛋白(N-cadherin)及波形蛋白(vimentin),评价上皮间质转化(EMT)发生情况。结果GEO数据资料分析表明,SNHG1在肺癌组织中明显高表达,且与患者的总生存期呈负相关;过表达SNHG1后NSCLC细胞的迁移及侵袭能力明显增加,上皮标志蛋白降低,间质标志蛋白增加;而干扰SNHG1则得到相反的结果。结论LncRNA SNHG1可通过EMT促进NSCLC细胞迁移和侵袭,提示LncRNA SNHG1有望成为NSCLC治疗的新靶点。
Objective To investigate the effect of long-chain non-coding RNA(LncRNA)SNHG1 on the migration and invasion of non-small cell lung cancer(NSCLC)cells.Methods The expression of SNHG1 in the lung cancer tissues and normal tissues was analyzed by using the datasets in the GEO database.The relationship between the expression of SNHG1 and the overall survival of lung cancer patients was analyzed.The expression of SNHG1 was overexpressed and interfered in NSCLC cells,the expression efficiency of SNHG1 was detected by qRT-PCR,and the effect of SNHG1 on the migration and invasion of NSCLC cells were detected by the cell scratch assay and Transwell migration and invasion assay respectively.Western blot(WB)was used to detect E-cadherin,N-cadherin and vimentin,and the occurrence of epithelial-mesenchymal transition(EMT)was evaluated.Results The results of the GEO data analysis showed that SNHG1 was highly expressed in the lung cancer tissues and negatively correlated with the overall survival of patients.After the overexpression of SNHG1,the migration and invasion ability of NSCLC cells significantly increased,the epithelial marker protein decreased and the interstitial marker protein increased.The interference of SNHG1 resulted in the opposite result.Conclusion LncRNA SNHG1 can promote the migration and invasion of NSCLC cells by EMT,which suggests that LncRNA SNHG1 is expected to be a new target for the treatment of non-small cell lung cancer.
